Abstract/Results: RESULTS:
We used event related potentials (ERPs) and sLORETA to determine how the management of processing cost in pronoun interpretation is affected by age (young vs. elderly) and by the presence of a cue for pragmatic inferencing. The syntactic relation between an explicit pronoun and its antecedent was manipulated, yielding two types of sentences (Type 1: syntactic (cheap) or discourse (costly) processing routes available; violation of a conversational convention vs. Type 2: exclusive availability of the discourse route; conformity to conversational conventions). For the young adults a Late Positivity and selective activation of right BA9 and BA31 emerged in type 1 sentences, indexing the effort required to compute the pragmatic inference cued by the violation of a conversational convention (use of an explicit pronoun in type 1 sentences, where a null/silent would be more likely). This effect is absent in older participants, suggesting that, in their case, the processor fails to initially capture the oddity of the explicit pronoun, which is therefore conveniently processed at the cheaper syntactic level, but rendering the identity of its original form unavailable for later pragmatic inferential processing. Thus, older participants, unlike the young, recruit different processing routes for each sentence type, as demonstrated by an N400 and selective activation of right BA18 for type 2 sentences, in which only the discourse route is available. This N400 is absent in younger participants, since the clash between the expected null pronoun and the explicit one occurring in type 1 sentences leads the processor to shift resolution to the discourse level, where the unexpected form will remain accessible to the pragmatic module.

Elderly patients may present with prominent cognitive complaints and have performances in neuropsychological tests within the normal range for the age and education, and thus do not fulfill the criteria for mild cognitive impairment (MCI). There is insufficient evidence to support the clinical decision in these cases ("pre-MCI"). Forty-three subjects, 11 controls, 15 "pre-MCI," and 17 MCI, were followed for about three and half years with neuropsychological testing and magnetic resonance imaging including volumetric measurements of the hippocampus and amygdala. Two of the "pre-MCI" subjects suffered cognitive and functional deterioration and were diagnosed with dementia. Although the "pre-MCI" subjects as a group had no significant deterioration in neuropsychological tests, they suffered a decline in the total hippocampal volume (P=0.04) along the follow-up time. In contrast, all control subjects remained stable and had no volumetric decreases. As expected, MCI patients underwent significant deterioration in several neuropsychological tests, often progressed to Alzheimer's disease, and showed decreases both in total hippocampal and amygdalar volumes. Elderly people presenting with cognitive complaints may be in an initial phase of a degenerative disorder and should be followed clinically, even if they have normal neuropsychological tests.

Abstract/Results: ABSTRACT:
The nature of visual impairments in Alzheimer's disease (AD) and their relation with other cognitive deficits remains highly debated. We asked whether independent visual deficits are present in AD and amnestic forms of mild cognitive impairment (MCI) in the absence of other comorbidities by performing a hierarchical analysis of low-level and high-level visual function in MCI and AD. Since parietal structures are a frequent pathophysiological target in AD and subserve 3D vision driven by motion cues, we hypothesized that the parietal visual dorsal stream function is predominantly affected in these conditions. We used a novel 3D task combining three critical variables to challenge parietal function: 3D motion coherence of objects of unknown orientation, with constrained temporal integration of these cues. Groups of amnestic MCI (n = 20), AD (n = 19), and matched controls (n = 20) were studied. Low-level visual function was assessed using psychophysical contrast sensitivity tests probing the magnocellular, parvocellular, and koniocellular pathways. We probed visual ventral stream function using the Benton Face Recognition task. We have found hierarchical visual impairment in AD, independently of neuropsychological deficits, in particular in the novel parietal 3D task, which was selectively affected in MCI. Integration of local motion cues into 3D objects was specifically and most strongly impaired in AD and MCI, especially when 3D motion was unpredictable, with variable orientation and short-lived in space and time. In sum, specific early dorsal stream visual impairment occurs independently of ventral stream, low-level visual and neuropsychological deficits, in amnestic types of MCI and AD.

The BIAL Foundation was created in 1994 by Laboratórios BIAL in conjunction with the Council of Rectors of Portuguese Universities. BIAL’s Foundation mission is to foster the scientific study of Man from both the physical and spiritual perspectives.
Along the years the BIAL Foundation has developed an important relationship with the scientific community, first in Portugal and after worldwide. Today it is an institution of reference which aims to stimulate new researches that may help people, promote more health and contribute to new milestones to gain access to knowledge.
Among its activities the BIAL Foundation manages the BIAL Award, created in 1984, one of the most important awards in the Health field in Europe. The BIAL Award rewards both the basic and the clinical research distinguishing works of major impact in medical research.
The BIAL Foundation also assigns Scientific Research Scholarships for the study of neurophysiological and mental health in people, arousing the interest of researchers in the areas of Psychophysiology and Parapsychology.
To date the BIAL Foundation has supported 461 projects, more than 1000 researchers, with research groups in twenty-seven countries, resulting, until April 2013, in about 600 full papers, out of which 172 published in indexed international journals with an average impact factor of 3.6 and a substantial number of citations (1665).
Since 1996 the BIAL Foundation organizes the Symposia entitled "Behind and Beyond the Brain", a Forum that gathers well renowned neurosciences speakers and the BIAL Foundation Fellows which are spread around the world.
Classified as an institution of public utility, the BIAL Foundation includes among its patrons the Portuguese President, the Portuguese Universities Rectors' Council and the Portuguese Medical Association.

Abstract/Results: ABSTRACT:
OBJECTIVE:
Mild cognitive impairment (MCI) is thought to represent a transitional stage between normal aging and early dementia, particularly Alzheimer’s disease (AD). Visual impairments have been reported in AD, consistently with the presence of cortical degeneration in visual areas. We aimed to investigate whether such deficits are already present at the stage of MCI.
METHODS:
Groups of amnestic MCI (N=20), AD (N=20) and matched controls (N=20) were studied. Patients were recruited following diagnosis through neurological and neuropsychological assessment, using Petersen’s classification criteria for MCI. All participants provided informed consent and underwent ophthalmologic examination to exclude any relevant retinal pathology.
Cognitive function was assessed using a neuropsychological battery, consisting of: WAIS III - Vocabulary, Digit Span and Digit Symbol; Rey Auditory Verbal Learning Test (AVLT); Benton Visual Retention Test and Benton Face Recognition Test. Visual function was further assessed using psychophysical tests: Frequency Doubling Test; Cambridge Colour Test; Structure From Motion (SFM) perception and visual search.
RESULTS:
Results: Statistical analysis of the data showed significant effects of group in all tests. Post-hoc comparison between groups revealed significant impairment of AD patients relative to controls in all tests, while MCI patients were impaired only in AVLT recall, Digit Symbol task, SFM discrimination threshold and visual search reaction times. In all cases, MCI patients were significantly less impaired than AD patients.
DISCUSSION:
Our results confirm the presence of severe visual function deficits in AD, by using objective psychophysical tests of contrast sensitivity, colour perception, structure from motion and face perception. In MCI patients, specific deficits were found for SFM and visual search, suggesting that parietal function becomes affected early in the course of disease, while occipital cortex degeneration probably occurs only at a later stage of dementia. Overall, our results indicate a dissociation between dorsal and ventral visual stream deficits in amnestic types of dementia.

Using psychophysical approaches, we aim to compare visual impairment in Dementia with Lewy Bodies (DLB) and Alzheimer´s Disease (AD). Twenty DLB patients, 20 AD patients and 20 controls were recruited. Participants without retinal pathology were assessed with: Local Speed Discrimination, Structure-From-Motion perception and a Contrast Sensitivity (CS) test under eye movement control. Both clinical groups had impaired performance in all visual tasks. The consistent difference between groups was that DLB patients were less able to inhibit loss of fixation in the CS task, in the presence of distracting pre-cues. Occipital networks involved in contrast sensitivity and motion perception are equally affected in both diseases. DLB patients showed a significant inability to control fixation, a biomarker that may differentiate these groups.

Herein, we focused on the study of the choroid plexus (CP), which constitutes the blood–cerebrospinal fluid barrier, in aging and in AD. Specifically, we used the PDGFB-APPSwInd (J20) transgenic mouse model of AD, which presents early memory decline and progressive Aß accumulation, and littermate age-matched wild-type (WT) mice, to characterize the CP transcriptome at 3, 5–6 and 11–12 months of age. The most striking observation was that the CP of J20 mice displayed an overall overexpression of type I interferon (IFN) response genes at all ages. Moreover, J20 mice presented a high expression of type II IFN genes in the CP at 3 months, which became lower than WT at 5–6 and 11–12 months. Importantly, along with a marked memory impairment and increased glial activation, J20 mice also presented a similar overexpression of type I IFN genes in the dorsal hippocampus at 3 months. Altogether, these findings provide new insights on a possible interplay between type I and II IFN responses in AD and point to IFNs as targets for modulation in cognitive decline.

Alzheimer’s disease (AD) is a neurodegenerative disease characterized by a marked decline in cognition and memory function. One of the major pathological hallmarks of AD is the progressive accumulation and deposition of amyloid beta (Aß) peptides in the brain. Importantly, increased Aß toxicity and pathology is accompanied by alterations in parallel mechanisms and responses that regulate brain homeostasis and function. Increasing evidence highlights the essential role of neuroinflammatory and immune-related molecules, including those produced at the brain barriers, on brain immune surveillance, decreased Aß clearance from the brain and increased brain cell dysfunction and pathology in AD. Therefore, understanding the response at the brain barriers may unravel novel pathways of relevance for the pathophysiology of AD. Alzheimer's disease (AD) is a neurodegenerative disorder whose main symptom is the progressive loss of cognitive abilities with age. One of the main pathological hallmarks of AD is the increased brain production, accumulation and aggregation of amyloid beta (Aß) peptides that lead to the formation of extracellular senile plaques. Familial AD, characterized by an early-onset, is attributed to mutations in specific genes involved in the amyloidogenic pathway, such as the gene encoding for amyloid precursor protein (APP). Risk for developing sporadic or late-onset AD was also attributed to diverse genetic abnormalities. Still, there is much to know about the mechanisms and biological agents underlying the genesis and progression of both forms of AD. Recent advances regarding the amyloid cascade hypothesis suggest that alterations in the processing of APP and Aß accumulation in the brain are a consequence of age-associated changes that take place at the level of the barriers of the brain, which precede neuronal and synaptic dysfunction, gliosis, cell death and overall brain pathology and atrophy. The less studied blood-cerebrospinal fluid (CSF) barrier is formed by the choroid plexus (CP) epithelial cells that produce most of the main constituents of the CSF. Importantly, the CP is also involved in the excretion of Aß out of the brain through different pathways. Herein we show that relevant changes occur at the level of the blood-CSF barrier, particularly regarding the CP transcriptome, along the process of aging of PDGFB-APPSwInd mice and of age-matched wild-type littermate controls. These changes might be associated with an impairment of the CP and of the blood-CSF-barrier, which is thought to occur with aging, as well as a less efficient clearance of Aß peptides from the brain in AD.

Alzheimer's disease (AD) is the most common form of dementia, whose prevalence is growing along with the increased life expectancy. Although the accumulation and deposition of amyloid beta (Aß) peptides in the brain is viewed as one of the pathological hallmarks of AD and underlies, at least in part, brain cell dysfunction and behavior alterations, the etiology of this neurodegenerative disease is still poorly understood. Noticeably, increased amyloid load is accompanied by marked inflammatory alterations, both at the level of the brain parenchyma and at the barriers of the brain. However, it is debatable whether the neuroinflammation observed in aging and in AD, together with alterations in the peripheral immune system, are responsible for increased amyloidogenesis, decreased clearance of Aß out of the brain and/or the marked deficits in memory and cognition manifested by AD patients. Herein, we scrutinize some important traits of the pathophysiology of aging and AD, focusing on the interplay between the amyloidogenic pathway, neuroinflammation and the peripheral immune system.

Abstract/Results: ABSTRACT:
Alzheimer’s disease (AD) is the most prevalent age-related dementia affecting millions of people worldwide. Its main pathological hallmark feature is the formation of insoluble protein deposits of amyloid-ß and hyperphosphorylated tau protein into extracellular plaques and intracellular neurofibrillary tangles, respectively. Many of the mechanistic details of this process remain unknown, but a well-established consequence of protein aggregation is synapse dysfunction and neuronal loss in the AD brain. Different pathways including mitochondrial dysfunction, oxidative stress, inflammation, and metal metabolism have been suggested to be implicated in this process. In particular, a body of evidence suggests that neuronal metal ions such as copper, zinc, and iron play important roles in brain function in health and disease states and altered homeostasis and distribution as a common feature across different neurodegenerative diseases and aging. In this focused review, we overview neuronal proteins that are involved in AD and whose metal binding properties may underlie important biochemical and regulatory processes occurring in the brain during the AD pathophysiological process.

S100B protein is one of the most abundant pro-inflammatory proteins which is chronically up-regulated in Alzheimer’s disease (AD) and is found within senile plaques [1]. Moreover, S100B is engaged in several processes linked with AD: regulates APP levels and generation of Aß peptides, interferes with tau protein phosphorylation and is involved in several AD-related signaling pathways. Recently we found that S100B binds to amyloid-ß peptide (Aß42) monomers and fibrils and acts as a suppressor of amyloid-ß peptide aggregation and toxicity [2]. Our finding raised the need to develop molecules that target S100B as a novel approach to ameliorate AD neurodegeneration. To fill this gap, we developed and purified a library of 24 single domain antibodies (nanobodies) targeting S100B protein in order to develop biological tools to stabilize the S100B:Aß42 complex. In most cases, the nanobody:S100B interaction occurs spontaneously, being favored by enthalpic forces. As determined by microscale thermophoresis (MST) and isothermal titration calorimetry (ITC), the binding affinity of nanobody:S100B interactions varies from high to low affinity (Kd=6-3000 nM) and the stoichiometry varies from 1 to 2 nanobodies per S100B protein. We found one nanobody that potentiates the inhibitory effect of S100B over Aß42 aggregation, promoting a strong suppression of Aß42 fibrillation, probably by stabilizing the S100B:Aß42 interaction. These newly developed nanobodies open new avenues to study other protein-protein interactions involving S100B on relevant disease processes.

Patients with Alzheimer's disease have an impairment of inhibitory control for reasons that are currently unclear. Using an eye-tracking task (the gap-overlap paradigm), we examined whether the uncorrected errors relate to the task of attentional disengagement in preparation for action. Alternatively, the difficulty in correcting for errors may be caused by the working memory representation of the task. A major aim of this study was to distinguish between the effects of healthy aging and neurodegenerative disease on the voluntary control of saccadic eye movements. Using the antisaccade task (AST) and pro-saccade task (PST) with the 'gap' and 'overlap' procedures, we obtained detailed eye-tracking measures in patients, with 18 patients with probable Alzheimer's disease, 25 patients with Parkinson's disease and 17 healthy young and 18 old participants. Uncorrected errors in the AST were selectively increased in Alzheimer's disease, but not in Parkinson's disease compared to the control groups. These uncorrected errors were strongly correlated with spatial working memory. There was an increase in the saccade reaction times to targets that were presented simultaneously with the fixation stimulus, compared to the removal of fixation. This 'gap' effect (i.e. overlap-gap) saccade reaction time was elevated in the older groups compared to young group, which yielded a strong effect of aging and no specific effect of neurodegenerative disease. Healthy aging, rather than neurodegenerative disease, accounted for the increase in the saccade reaction times to the target that are presented simultaneously with a fixation stimulus. These results suggest that the impairment of inhibitory control in the AST may provide a convenient and putative mark of working memory dysfunction in Alzheimer's disease.

Alzheimer's disease (AD) is characterized by different clinical entities. Although AD phenotypes share a common molecular substrate (i.e., amyloid beta and tau accumulation), several clinicopathological differences exist. Brain functional networks might provide a macro-scale scaffolding to explain this heterogeneity. In this review, we summarize the evidence linking different large-scale functional network abnormalities to distinct AD phenotypes. Specifically, executive deficits in early-onset AD link with the dysfunction of networks that support sustained attention and executive functions. Posterior cortical atrophy relates to the breakdown of visual and dorsal attentional circuits, while the primary progressive aphasia variant of AD may be associated with the dysfunction of the left-lateralized language network. Additionally, network abnormalities might provide in vivo signatures for distinguishing proteinopathies that mimic AD, such as TAR DNA binding protein 43 related pathologies. These network differences vis-a-vis clinical syndromes are more evident in the earliest stage of AD. Finally, we discuss how these findings might pave the way for new tailored interventions targeting the most vulnerable brain circuit at the optimal time window to maximize clinical benefits.

he study of pollutant effects is extremely important to address the epochal challenges we are facing, where world populations are increasingly moving from rural to urban centers, revolutionizing our world into an urban world. These transformations will exacerbate pollution, thus highlighting the necessity to unravel its effect on human health. Epidemiological studies have reported that pollution increases the risk of neurological diseases, with growing evidence on the risk of neurodegenerative disorders. Air pollution and water pollutants are the main chemicals driving this risk. These chemicals can promote inflammation, acting in synergy with genotype vulnerability. However, the biological underpinnings of this association are unknown. In this review, we focus on the link between pollution and brain network connectivity at the macro-scale level. We provide an updated overview of epidemiological findings and studies investigating brain network changes associated with pollution exposure, and discuss the mechanistic insights of pollution-induced brain changes through neural networks. We explain, in detail, the pollutome-connectome axis that might provide the functional substrate for pollution-induced processes leading to cognitive impairment and neurodegeneration. We describe this model within the framework of two pollutants, air pollution, a widely recognized threat, and polyfluoroalkyl substances, a large class of synthetic chemicals which are currently emerging as new neurotoxic source.
