Final report - Event-related potentials of temperament traits in ADHD and conduct disorder

Abstract/Results: RESULTS:
The aim of this study was to understand brain function involved in symptoms and temperament traits of ADHD and conduct disorder. The project includes 80 adolescent boys into the study (30 healthy individuals, 30 boys with ADHD, 20 boys with conduct disorder). We also made use of an international database of 245 children and adolescents to investigate maturational changes in brain function.
A structured clinical interview was used to diagnose developmental problems including Attention Deficit and Hyperactivity Disorder (ADHD), oppositional defiant disorder (ODD) and Conduct Disorder (CD). A number of techniques were used to measure brain function including electroencephalography, event-related potentials and neuropsychology measures. Brain function assessment was performed using brain resource company techniques (www.brainresource.com). We also used several self and parent report instruments to measures symptoms and temperament traits, in particular we were interested in callous/unemotional traits, emotional intelligence, affective problems, pragmatic language impairment, hyperactivity, impulsivity, and aggression. Data from this project have been presented in poster and oral form at the Applied Neuroscience Conference, May 17-20, 2007. Two manuscripts have been submitted, to Developmental Neuropsychology and to The Journal of Child Psychology and Psychiatry. The first of these studies investigates sex differences in the ontogenetic development of N200 and P300 components of the event-related potential. The second observes
neurodevelopmental differences between healthy and behaviourally problematic adolescents, and investigates the relationship of involved mechanisms to clinical scores. Further publications from this dataset are in preparation.

Event-related potentials analysed in consecutive studies led us to the following findings: a) the perception of fear from faces is gated by selective attention at early latencies, whereas only after 480 ms was the perception of disgust modulated by attention allocation; b) the order of presentation of external and internal facial features influence the integration of facial components into face gestalts, so that the external features may be especially relevant for initial object categorization while internal features would be more closely related to subsequent configural mechanisms; c) the verbal N400 wave was distinguished from its functional analogue during the processing of faces in a contextual preactivation task, which showed a predominantly occipital localization and differentiated neural generators; d) N400-like components elicited in a face-feature matching task revealed a larger and longer-lasting N400 effect with the prior presentation of internal features around 300-400 ms post-stimulus, involving a greater activation of frontal and left temporal brain areas; e) both explicit and implicit processing of famous faces resulted in an enhanced N250 whereas explicit processing was specifically associated with earlier N400 and P600, related to an increased activity within brain areas involved in identity processing around 250 and 450 ms; and f) the processing of familiar vs. unfamiliar faces in the face-feature matching task revealed that the so-called “internal features advantage” relies on the use of stored face-identity-related information around 300-600 ms, involving an incremented activity elicited by familiar stimuli in both posterior (ventral occipitotemporal) and more anterior (parahippocampal and orbitofrontal) brain regions.

In 1994 the BIAL Foundation launched a programme of science research grants with the aim of encouraging the research into Man’s physical and mental processes, namely in fields still largely unexplored but which warrant further scientific analysis, as Psychophysiology and Parapsychology.
Since its launch, applications to the BIAL grants have been increasing. Up to now 461 projects have been supported, involving more than 1000 researchers from 27 countries.
The approved applications have benefited from grants in amounts comprised between €5,000 and €50, 000. The amount to be granted is fixed by the Scientific board according to the needs of each project.

Abstract/Results: RESULTS:
We used event related potentials (ERPs) and sLORETA to determine how the management of processing cost in pronoun interpretation is affected by age (young vs. elderly) and by the presence of a cue for pragmatic inferencing. The syntactic relation between an explicit pronoun and its antecedent was manipulated, yielding two types of sentences (Type 1: syntactic (cheap) or discourse (costly) processing routes available; violation of a conversational convention vs. Type 2: exclusive availability of the discourse route; conformity to conversational conventions). For the young adults a Late Positivity and selective activation of right BA9 and BA31 emerged in type 1 sentences, indexing the effort required to compute the pragmatic inference cued by the violation of a conversational convention (use of an explicit pronoun in type 1 sentences, where a null/silent would be more likely). This effect is absent in older participants, suggesting that, in their case, the processor fails to initially capture the oddity of the explicit pronoun, which is therefore conveniently processed at the cheaper syntactic level, but rendering the identity of its original form unavailable for later pragmatic inferential processing. Thus, older participants, unlike the young, recruit different processing routes for each sentence type, as demonstrated by an N400 and selective activation of right BA18 for type 2 sentences, in which only the discourse route is available. This N400 is absent in younger participants, since the clash between the expected null pronoun and the explicit one occurring in type 1 sentences leads the processor to shift resolution to the discourse level, where the unexpected form will remain accessible to the pragmatic module.

Little is known about how sex influences functional brain maturation. The current study investigated sex differences in the maturation of event-related potential (ERP) amplitudes during an auditory oddball task (N = 170; age = 6–17 years). Performance improved with age. N200 amplitude declined with age: parietal sites showed earlier development than temporal and frontal locations. Girls showed greater bilateral frontal P300 amplitude development, approaching the higher values observed in boys during childhood. After controlling for age, right frontal P300 amplitude was associated with reaction time in girls. The findings demonstrate sex differences in ERP maturation in line with behavioral and neuroimaging studies.

General anesthesia reversibly alters consciousness, without shutting down the brain globally. Depending on the anesthetic agent and dose, it may produce different consciousness states including a complete absence of subjective experience (unconsciousness), a conscious experience without perception of the environment (disconnected consciousness, like during dreaming), or episodes of oriented consciousness with awareness of the environment (connected consciousness). Each consciousness state may potentially be followed by explicit or implicit memories after the procedure. In this respect, anesthesia can be considered as a proxy to explore consciousness. During the recent years, progress in the exploration of brain function has allowed a better understanding of the neural correlates of consciousness, and of their alterations during anesthesia. Several changes in functional and effective between-region brain connectivity, consciousness network topology, and spatio-temporal dynamics of between-region interactions have been evidenced during anesthesia. Despite a set of effects that are common to many anesthetic agents, it is still uneasy to draw a comprehensive picture of the precise cascades during general anesthesia. Several questions remain unsolved, including the exact identification of the neural substrate of consciousness and its components, the detection of specific consciousness states in unresponsive patients and their associated memory processes, the processing of sensory information during anesthesia, the pharmacodynamic interactions between anesthetic agents, the direction-dependent hysteresis phenomenon during the transitions between consciousness states, the mechanisms of cognitive alterations that follow an anesthetic procedure, the identification of an eventual unitary mechanism of anesthesia-induced alteration of consciousness, the relationship between network effects and the biochemical or sleep-wake cycle targets of anesthetic agents, as well as the vast between-studies variations in dose and administration mode, leading to difficulties in between-studies comparisons. In this narrative review, we draw the picture of the current state of knowledge in anesthesia-induced unconsciousness, from insights gathered on propofol, halogenated vapors, ketamine, dexmedetomidine, benzodiazepines and xenon. We also describe how anesthesia can help understanding consciousness, we develop the above-mentioned unresolved questions, and propose tracks for future research.

Neuroscientists agree on the value of locating the source of consciousness within the brain. Anaesthesiologists are no exception, and have their own operational definition of consciousness based on phenomenological observations during anaesthesia. The full functional correlates of consciousness are yet to be precisely identified, however rapidly evolving progress in this scientific domain has yielded several theories that attempt to model the generation of consciousness. They have received variable support from experimental observations, including those involving anaesthesia and its ability to reversibly modulate different aspects of consciousness. Aside from the interest in a better understanding of the mechanisms of consciousness, exploring the functional tenets of the phenomenological consciousness states of general anaesthesia has the potential to ultimately improve patient management. It could facilitate the design of specific monitoring devices and approaches, aiming at reliably detecting each of the possible states of consciousness during an anaesthetic procedure, including total absence of mental content (unconsciousness), and internal awareness (sensation of self and internal thoughts) with or without conscious perception of the environment (connected or disconnected consciousness, respectively). Indeed, it must be noted that unresponsiveness is not sufficient to infer absence of connectedness or even absence of consciousness. This narrative review presents the current knowledge in this field from a system-level, underlining the contribution of anaesthesia studies in supporting theories of consciousness, and proposing directions for future research.

The BIAL Foundation was created in 1994 by Laboratórios BIAL in conjunction with the Council of Rectors of Portuguese Universities. BIAL’s Foundation mission is to foster the scientific study of Man from both the physical and spiritual perspectives.
Along the years the BIAL Foundation has developed an important relationship with the scientific community, first in Portugal and after worldwide. Today it is an institution of reference which aims to stimulate new researches that may help people, promote more health and contribute to new milestones to gain access to knowledge.
Among its activities the BIAL Foundation manages the BIAL Award, created in 1984, one of the most important awards in the Health field in Europe. The BIAL Award rewards both the basic and the clinical research distinguishing works of major impact in medical research.
The BIAL Foundation also assigns Scientific Research Scholarships for the study of neurophysiological and mental health in people, arousing the interest of researchers in the areas of Psychophysiology and Parapsychology.
To date the BIAL Foundation has supported 461 projects, more than 1000 researchers, with research groups in twenty-seven countries, resulting, until April 2013, in about 600 full papers, out of which 172 published in indexed international journals with an average impact factor of 3.6 and a substantial number of citations (1665).
Since 1996 the BIAL Foundation organizes the Symposia entitled "Behind and Beyond the Brain", a Forum that gathers well renowned neurosciences speakers and the BIAL Foundation Fellows which are spread around the world.
Classified as an institution of public utility, the BIAL Foundation includes among its patrons the Portuguese President, the Portuguese Universities Rectors' Council and the Portuguese Medical Association.

A fundamental challenge in neuroscience is accurately defining brain states and predicting how and where to perturb the brain to force a transition. Here, we investigated resting-state fMRI data of patients suffering from disorders of consciousness (DoC) after coma (minimally conscious and unresponsive wakefulness states) and healthy controls. We applied model-free and model-based approaches to help elucidate the underlying brain mechanisms of patients with DoC. The model-free approach allowed us to characterize brain states in DoC and healthy controls as a probabilistic metastable substate (PMS) space. The PMS of each group was defined by a repertoire of unique patterns (i.e., metastable substates) with different probabilities of occurrence. In the model-based approach, we adjusted the PMS of each DoC group to a causal whole-brain model. This allowed us to explore optimal strategies for promoting transitions by applying off-line in silico probing. Furthermore, this approach enabled us to evaluate the impact of local perturbations in terms of their global effects and sensitivity to stimulation, which is a model-based biomarker providing a deeper understanding of the mechanisms underlying DoC. Our results show that transitions were obtained in a synchronous protocol, in which the somatomotor network, thalamus, precuneus and insula were the most sensitive areas to perturbation. This motivates further work to continue understanding brain function and treatments of disorders of consciousness.
