A great challenge in neuroscience is understanding how activity in the brain gives rise to behavior. The zebrafish is an ideal vertebrate model to address this challenge, thanks to the capacity, at the larval stage, for precise behavioral measurements, genetic manipulations, and recording and manipulation of neural activity noninvasively and at single-neuron resolution throughout the whole brain. These techniques are being further developed for application in freely moving animals and juvenile stages to study more complex behaviors including learning, decision making, and social interactions.Wereview some of the approaches that have been used to study the behavior of zebrafish and point to opportunities and challenges that lie ahead.

Abstract/Results: ABSTRACT:
“Wanting”, a component of reward processing, is a motivational property that guides decision making in goal-oriented behavior. This includes behavior aiming at supporting relational bonds, even at the group level. Accordingly, group belongingness works as this motivational property, which is fundamentally different from romantic or maternal love. While primary rewards (or learned associations, such as money) have been largely used to study the conceptual framework associated with “wanting,” other cues triggering behavior, such as passionate motives, are less well-studied. We investigated the neural correlates of value estimation of a passion-driven incentive in neuropsychologically defined football fans. We asked the participants (n = 57) to compute the value of football tickets (the cues that trigger passionate behavior in this “tribal love” context). The trials were all different, comprising tickets for different matches. The participants had no restrictions on the amount to be introduced. This enabled a parametric functional magnetic resonance imaging design based on the explicit estimated value given by the participants in a trial-by-trial approach. Using a whole-brain approach (to prevent biased focus on value-related regions), only the activity in the ventral caudate and left anterior insula showed a critical relationship with the reported value. Higher normalized values led to more activity in the striatum and left insula. The parametric map shows that these regions encode the magnitude of incentive by indexing self-relevant value. Other regions were involved in value computation, such as the ventromedial prefrontal cortex, lateral orbitofrontal cortex, and dorsolateral prefrontal cortex, but did not exhibit parametric patterns. The involvement of the nucleus accumbens in value estimation was only found in region of interest -based analysis, which emphasizes the role of the ventral caudate for the presently studied social “reinforcer” cue.

Abstract/Results: ABSTRACT:
Goal-directed behavior requires integrating sensory information with prior knowledge about the environment. Behavioral biases that arise from these priors could increase positive outcomes when the priors match the true structure of the environment, but mismatches also happen frequently and could cause unfavorable outcomes. Biases that reduce gains and fail to vanish with training indicate fundamental suboptimalities arising from ingrained heuristics of the brain. Here, we report systematic, gain-reducing choice biases in highly trained monkeys performing a motion direction discrimination task where only the current stimulus is behaviorally relevant. The monkey’s bias fluctuated at two distinct time scales: slow, spanning tens to hundreds of trials, and fast, arising from choices and outcomes of the most recent trials. Our findings enabled single trial prediction of biases, which influenced the choice especially on trials with weak stimuli. The pre-stimulus activity of neuronal ensembles in the monkey prearcuate gyrus represented these biases as an offset along the decision axis in the state space. This offset persisted throughout the stimulus viewing period, when sensory information was integrated, leading to a biased choice. The pre-stimulus representation of history-dependent bias was functionally indistinguishable from the neural representation of upcoming choice before stimulus onset, validating our model of single-trial biases and suggesting that pre-stimulus representation of choice could be fully defined by biases inferred from behavioral history. Our results indicate that the prearcuate gyrus reflects intrinsic heuristics that compute bias signals, as well as the mechanisms that integrate them into the oculomotor decision-making process.

Daily, individuals select actions based on cost-benefit to allocate resources into goal-directed actions. Different brain regions coordinate this complex decision, including the nucleus accumbens (NAc), anterior cingulate cortex (ACC), and ventral tegmental area (VTA). In utero exposure to synthetic glucocorticoids (iuGC), such as dexamethasone, triggers prominent motivation deficits but the impact of this exposure in the ACC-NAc and/or ACC-VTA circuits is unknown. Here, we show that iuGC exposure causes decreased motivation for natural rewards (food) and impaired effort-based decision-making. Importantly, reduced neuronal activation (number of c-fos+ neurons) was observed in the NAc core and ACC of iuGC rats in comparison to CTR rats after performing the effort-based decision-making task. In addition, iuGC treatment led to increased NAc and ACC basal neuronal activity. Electrophysiological recordings during optogenetic modulation of ACC terminals in the NAc revealed that the ACC-NAc circuit is dysfunctional in iuGC animals. These data suggest that iuGC animals present motivational and effort-based decision-making deficits that can be associated with the observed ACC-NAc dysfunction.

Redefining the decisional components of motor responses: Evidence from lexical and object decision tasks

Abstract/Results: ABSTRACT:
Models of decision making focusing on two-alternative choices have classically described motor-response execution as a nondecisional stage that serially follows the termination of decision processes. Recent evidence, however, points toward a more continuous transition between decision and motor processes. We investigated this transition in two lexical decisions and one object decision task. By recording the electromyographic (EMG) signal associated with the muscle responsible for the manual responses (i.e., button press), we partitioned single-trial reaction times into premotor (the time elapsing from stimulus onset until the onset of the EMG burst) and motor times (the time elapsing from the onset of the EMG burst and the button press), with the latter measuring response execution. Responses were slower for pseudowords and pseudo-objects compared to words and real objects. Importantly, these effects were reliable even at the level of motor time measures. Differently, despite the reliable effect at the level of reaction times and premotor times, there was no difference in motor times between high- and low-frequency words. Although these results, in line with recent evidence, challenge a purely noncognitive characterization of motor-response execution, they further suggest that motor times may selectively capture specific decisional components, which we identify with late-occurring verification and/or control mechanisms.

ABSTRACT:
Background
Prominent decision-making models assume a serial transition from cognition to action. Decisions are envisaged as evidence accumulating towards one of the alternatives, until reaching an action-triggering boundary: Motor responses are not part of the decision process.
Aims
The project questioned the notion of a functional segregation between cognition and action, and attempted to identify the neurocognitive underpinnings of a continuous processing stream mapping decisions to motor responses.
Method
The project focused on lexical decision experiments with manual button-press responses. The first study used electromyographic (EMG) traces to measure response durations. The second study co-registered EMG and electroencephalographic (EEG) signals to track the propagation of decisions across different stages of the motor hierarchy, including long-range interactions indexed by corticomuscular coherence (CMC).
Results
The first study revealed a selective propagation of decisional effects to response duration, which was possibly modulated by verification processes for items with no representation in memory. The second study highlighted a discontinuity in the percolation of decisional effects across the motor hierarchy, with the involvement of motor planning and response duration, but no modulations of motor programming and CMC.
Conclusions
These results question the assumption of a serial transition from decision to motor responses, and the notion of a single decisional variable propagating from decisional to motor stages. Differently, they foster the hypothesis of multiple decisional components reflected at different stages of motor control, pointing to verification process as a plausible decisional substrate of motor execution.

While neural networks approach human levels of perfor- mance in many complex tasks, they require much more training than humans. This may be because only humans can infer and apply generalizable principles from prior ex- perience (Lake, Ullman, Tenenbaum, & Gershman, 2017). However, the statistics that underlie the human learning process are poorly understood and hard to investigate in the large state spaces found in most complex tasks (van Opheusden & Ma, 2019). We thus designed a cog- nitive task whose potential solutions are few enough for subjects to densely sample policy space, but complex enough to compel intelligent search. We launched the game as a smartphone-based app (hexxed.io) to collect data from 10k human participants. We find that unlike re- inforcement learning agents (Deep-Q Networks ; DQNs), humans (1) search a highly restricted subset of the pol- icy space; (2) attempt even poor solutions many times before discarding them; (3) arrive at the optimal policy suddenly and unpredictably with a “leap of insight”. Our data suggest a “top-down” learning process by which hu- mans propose explanatory solutions which they replace only upon collecting sufficient evidence to the contrary, in contrast to the “bottom-up” learning of DQNs that as- sociates states with rewarding actions
