In 1994 the BIAL Foundation launched a programme of science research grants with the aim of encouraging the research into Man’s physical and mental processes, namely in fields still largely unexplored but which warrant further scientific analysis, as Psychophysiology and Parapsychology.
Since its launch, applications to the BIAL grants have been increasing. Up to now 461 projects have been supported, involving more than 1000 researchers from 27 countries.
The approved applications have benefited from grants in amounts comprised between €5,000 and €50, 000. The amount to be granted is fixed by the Scientific board according to the needs of each project.

Modulation of nociceptive transmission at the brainstem involves several neurochemical systems. The precise location and specific characteristics of nociceptive neurons activated in each system was never reported. In this study, the presence of GABA(B), mu-opioid, and neurokinin-1 (NK-1) receptors in brainstem nociceptive neurons was investigated by double-immunocytochemical detection of each receptor and noxious-evoked induction of the c-fos proto-oncogene. Noxious cutaneous mechanical stimulation significantly increased the proportions of neurons double-labelled for Fos and GABA(B) receptors in several brainstem regions, namely, the reticular formation of the caudal ventrolateral medulla (VLMlat and VLMrf), lateral reticular nucleus, spinal trigeminal nucleus, pars caudalis (Sp5C), nucleus of the solitary tract, dorsal reticular nucleus, ventral reticular nucleus, raphe obscurus nucleus and dorsal parabrachial nucleus (DPB). For mu-opioid receptors, the proportions of double-labelled neurons in noxious-stimulated animals were higher than in controls only in the VLMlat, VLMrf, Sp5C, DPB and A5 noradrenergic cell group. As for the NK-1 receptor, no significant differences were found between control and stimulated animals. According to these results, neurons expressing GABA(B), mu-opioid and NK-1 receptors at several pain control centres of the brainstem are differentially involved in processing nociceptive mechanical input. The data provide the definition of new supraspinal targets for selective modulation of nociceptive neurons in order to define better strategies of pain control.

Herpes Simplex Virus type 1 (HSV-1) vectors are known to inhibit nociceptive transmission at the spinal cord after peripheral applications. Similar approaches may also be useful when applied at the supraspinal pain control system as the system includes pronociceptive (facilitatory) components. We performed a study aimed to analyse the migration of HSV-1 along with the inhibition of pronociception from the medullary dorsal reticular nucleus (DRt), a major facilitatory component of the supraspinal pain control system. To study the migration, a HSV-1 vector expressing lacZ under control of the human cytomegalovirus (hCMV) promoter was injected in the DRt and the expression of ß-galactosidase (ß-gal) was detected at 2, 4, 7, 10 and 14 days. Numerous ß-gal-immunoreactive neurons were observed at the injection site until day 4, and at some of the brain areas projecting to the DRt until day 7. To block the pronociceptive effects of the DRt, a HSV-1 vector expressing the preproenkephalin transgene, under the control of hCMV promoter, was injected into the DRt. Behavioural evaluation was performed at the time-points referred above, using the paw withdrawal latency test to evaluate thermal nociceptive responses. Anti-hyperalgesic effects persisted during 4 days, decreasing after that time-point. The present study demonstrates that selective migration of HSV-1 should be considered in gene therapy strategies based on HSV-1 injections into the brain. The study also shows that it is possible to decrease pain facilitation from the brain using opioidergic inhibition of pronociceptive supraspinal areas.

Several authors have shown that the hippocampus responds to painful stimulation and suggested that prolonged painful conditions could lead to abnormal hippocampal functioning. The aim of the present study was to evaluate whether the induction of persistent peripheral neuropathic pain would affect basic hippocampal processing such as the spatial encoding performed by CA1 place cells. These place cells fire preferentially in a certain spatial position in the environment, and this spatial mapping remains stable across multiple experimental sessions even when the animal is removed from the testing environment. To address the effect of prolonged pain on the stability of place cell encoding, we chronically implanted arrays of electrodes in the CA1 hippocampal region of adult rats and recorded the multichannel neuronal activity during a simple food-reinforced alternation task in a U-shaped runway. The activity of place cells was followed over a 3-week period before and after the establishment of an animal model of neuropathy, spared nerve injury. Our results show that the nerve injury increased the number of place fields encoded per cell and the mapping size of the place fields. In addition, there was an increase in in-field coherence while the amount of spatial information content that a single spike conveyed about the animal location decreased over time. Other measures of spatial tuning (in-field firing rate, firing peak and number of spikes) were unchanged between the experimental groups. These results demonstrate that the functioning of spatial place cells is altered during neuropathic pain conditions.

Dynamics of Circadian Thalamocortical Flow of Information during a Peripheral Neuropathic Pain Condition

Abstract/Results: ABSTRACT:
It is known that the thalamocortical loop plays a crucial role in the encoding of sensory–discriminative features of painful stimuli. However, only a few studies have addressed the changes in thalamocortical dynamics that may occur after the onset of chronic pain. Our goal was to evaluate how the induction of chronic neuropathic pain affected the flow of information within the thalamocortical loop throughout the brain states of the sleep–wake cycle. To address this issue we recorded local field potentials (LFPs) – both before and after the establishment of neuropathic pain in awake freely moving adult rats chronically implanted with arrays of multielectrodes in the lateral thalamus and primary somatosensory cortex. Our results show that the neuropathic injury induced changes in the number of wake and slow-wave-sleep (SWS) state episodes, and especially in the total number of transitions between brain states. Moreover, partial directed coherence – analysis revealed that the amount of information flow between cortex and thalamus in neuropathic animals decreased significantly, indicating that the overall thalamic activity had less weight over the cortical activity. However, thalamocortical LFPs displayed higher phase-locking during awake and SWS episodes after the nerve lesion, suggesting faster transmission of relevant information along the thalamocortical loop. The observed changes are in agreement with the hypothesis of thalamic dysfunction after the onset of chronic pain, and may result from diminished inhibitory effect of the primary somatosensory cortex over the lateral thalamus.

Abstract/Results: RESULTS:
This research project aims at understanding the alterations that occur following the onset of prolonged and stressful pain conditions, in the connectivity of brain areas that process rewarding and aversive stimuli and in areas critical for learning and memory.
For achieving our goal we use a combination of novel decision-making and working-memory operant tasks together with state-of-the-art multielectrode neurophysiology recordings in awake freely moving animals. In a typical experiment we chronically implant 16-32 tungsten microwires in up to 4 brain areas.
The results of this project showed that:
a) the neuronal firing rate in the orbitofrontal cortex was correlated with the probability of choosing a low versus high-risk food reward in each trial, and that chronic pain reduced the fraction of risk-aversive neurons;
b) neuropathic pain induces an increase in the number of place fields encoded by hippocampal neurons;
c) chronic pain changes the circadian behavioral state leading to a disruption of sleep patterns, and that there was a large decrease in the functional connectivity between the somatosensory cortex and somatosensory lateral thalamus;
d) pain increases neuronal activity in the amygdala triggering a decrease in prefrontal activation and impairing decision-making;
e) pain induces an impairment of working memory performance, decrease in single neuron activity in the mPFC, and reduction in the frontohippocampal connectivity correlated with correct performance.
f) congenital lack of pain in Prrxl1 KO mice causes a behavioural and neurophysiological pattern of brain activity that is the inverse of what we observed in animals with either neuropathic or inflammatory chronic pain.

Abstract/Results: ABSTRACT:
Chronic pain patients commonly complain of working memory deficits, but the mechanisms and brain areas underlying this cognitive impairment remain elusive. The neuronal populations of the mPFC and dorsal CA1 (dCA1) are well known to form an interconnected neural circuit that is crucial for correct performance in spatial memory-dependent tasks. In this study, we investigated whether the functional connectivity between these two areas is affected by the onset of an animal model of peripheral neuropathic pain. To address this issue, we implanted two multichannel arrays of electrodes in the mPFC and dCA1 of rats and recorded the neuronal activity during a food-reinforced spatial working memory task in a reward-based alternate trajectory maze. Recordings were performed for 3 weeks, before and after the establishment of the spared nerve injury model of neuropathy. Our results show that the nerve lesion caused an impairment of working memory performance that is temporally associated with changes in the mPFC populational firing activity patterns when the animals navigated between decision points-when memory retention was most needed. Moreover, the activity of both recorded neuronal populations after the nerve injury increased their phase locking with respect to hippocampal theta rhythm. Finally, our data revealed that chronic pain reduces the overall amount of information flowing in the fronto-hippocampal circuit and induces the emergence of different oscillation patterns that are well correlated with the correct/incorrect performance of the animal on a trial-by-trial basis. The present results demonstrate that functional disturbances in the fronto-hippocampal connectivity are a relevant cause for pain-related working memory deficits.

It has been recently described that disruption of the neural mechanisms of emotion-based decision making occurs in both chronic pain patients and in animal models of pain; moreover, it also has been shown that chronic pain causes morphological and functional changes in the prefrontal cortex that may be crucial for this decision-making dysfunction. However, it is not known whether pain alone is capable of altering the neuronal encoding of decision exhibited by prefrontal neurons. We have previously shown that naïve animals have risk-averse performance in the rodent gambling task, whereas chronic pain animals reverse their choice preference and become risk prone. Using this paradigm, we chronically implanted arrays of multielectrodes and recorded from neuronal ensembles in the orbitofrontal cortex of freely moving animals performing 4 sessions of the rodent gambling task: 2 in control conditions and 2 after the onset of inflammatory pain induced by complete Freund’s adjuvant injection. Our results show that the instantaneous neuronal firing rate was correlated with the probability of choosing a specific lever in 62.5% of the neurons; however, although in the control sessions 61% of the neurons encoded the reward magnitude, after the pain onset only 16% of the neurons differentiated small from large rewards. Moreover, we found that the fraction of risk-sensitive neurons recorded in each session predicted the overall risk bias of the animal. Our data suggest that orbitofrontal cortex encoding of risk preference is compromised in chronic pain animals.

Title:
Comparison of anesthetic depth indexes based on thalamocortical local field potentials in rats

Abstract/Results: ABSTRACT:
BACKGROUND - Local field potentials may allow a more precise analysis of the brain electrical activity than the electroencephalogram. In this study, local field potentials were recorded in the thalamocortical axis of rats to (i) compare the performance of several indexes of anesthetic depth and (ii) investigate the existence of thalamocortical correlated or disrupted activity during isoflurane steady-state anesthesia.
METHODS - Five rats chronically implanted with microelectrodes were used to record local field potentials in the primary somatosensory cortex and ventroposterolateral thalamic nuclei at six periods: before induction of anesthesia; in the last 5 min of randomized 20-min steady-state end-tidal 0.8, 1.1, 1.4, and 1.7% isoflurane concentrations; and after recovery. The approximate entropy, the index of consciousness, the spectral edge frequency, and the permutation entropy were estimated using epochs of 8 s. A correction factor for burst suppression was applied to the spectral edge frequency and to the permutation entropy. The correlation between the derived indexes and the end-tidal isoflurane was calculated and compared for the two studied brain regions indexes. Coherence analysis was also performed.
RESULTS - The burst suppression-corrected permutation entropy showed the highest correlation with the end-tidal isoflurane concentration, and a high coherence was obtained between the two studied areas.
CONCLUSIONS - The permutation entropy corrected with the classic burst suppression ratio is a promising alternative to other indexes of anesthetic depth. Furthermore, high coherence level of activity exists between the somatosensory cortical and thalamic regions, even at deep isoflurane stages.

Abstract/Results: ABSTRACT:
Introduction: Burst suppression (BS) activity of the electroencephalogram (EEG) reflects deep anesthesia and has been used to help intra operative anesthetic depth monitoring. It appears as a transitional pattern between isoelectricity and high-frequency EEG and its quantification is normally based in the percentage of isoelectricity in EEG epochs¹. Tsallis entropy (TE) has been recently applied to quantify the presence of BS in the EEG of rats after cardiac arrest², but there is no data on this analysis during anesthesia. The use of a non-linear analysis based method (TE) could be a more practical alternative to the traditional BS ratio, as the first one does not need the set of an amplitude limit to consider the signal as isoelectric that may vary with amplitude and recording conditions. In the present study, the potential to use TE to quantify BS was accessed using a high resolution signal called local field potentials recorded in rats under isoflurane anesthesia. Methods: Five rats chronically implanted with microelectrodes were used to record local field potentials in the primary somatosensory at 6 periods: before induction of anesthesia; in the last 5 min of randomized 20 min steady state end-tidal 0.8%, 1.1%, 1.4% and 1.7% isoflurane concentrations (etISO) and after recovery. A 50 Hz notch filter was applied to the signal before analysis. Four epochs of 12 seconds (3000 data points per epoch) were selected from the last 5 min of each concentration step and analyzed: the classic burst suppression ratio (BSR)[start_en]00B9; and Tzallis entropy² were calculated. The Spearman Rank correlation coefficient was calculated between etISO and the BSR and between etISO and TE for each rat (24 pairs of data each rat). A paired sample t-test was used to compare the absolute values of the correlation coefficients of the BSR and the TE (n=5). Results: Figure 1 shows the absolute values for the correlation coefficients between etISO and BSR and between etISO and TE for the five rats.[figure1]The TE varied between 0,429 and 0.499 and decreased with increasing anesthetic dosages, resulting in negative correlation coefficients. The BSR had significant positive correlation coefficients, except in one animal in which the correlation coefficient was bellow 0.5. There were no significant differences between the correlation coefficients of the two indexes with etISO. However, TE had a higher mean of the correlation coefficients. Conclusions: Although the BSR is a very simple and fast method to quantify BS in the EEG, the use of a non-linear analysis method such as TE eliminates the need to perform visual inspection of the signal in order to pre-set the amplitude limit of isoelectricity. These results point out the potential of TE to be applied in practice. However, further investigation is required with extracranial signals.

Altered processing in orbitofrontal cortex activity in monoarthritic rats during a rodent gambling task

Abstract/Results: ABSTRACT:
The orbitofrontal cortex (OFC) is fundamental for the update of reward value in decision making tasks in humans and animals. Previous works in rats have shown that lesions of the OFC, as well as chronic pain lead to impairment in the Rodent Gambling Task akin to the Iowa Gambling Task (Bechara et al., 1994). Here we used multielectrode recordings to study orbitofrontal cortex activity in rats performing this task during control and chronic pain conditions. As expected, control animals developed preference for low risk choices while monoarthritic rats developed preference for high risk choices. Neuronal activity showed that control animals present a higher proportion of cells that differentially fire for high and low risk choices during the whole task, while monoarthritic animals only present this differential activity in the beginning of the task. Comparison of neuronal responses to rewards showed that control animals presented a higher proportion of cells with different firing rates for high and low risk rewards, but that the pain group presented an increased number of cells that differentially fired for rewarded and non rewarded trials in the low risk lever. These results show for the first time altered orbitofrontal cortex processing during decision-making deficits in chronic pain conditions and support the notion that chronic pain alters the reward/aversion system.

Withholding impulsive actions for short term gains to achieve longer term goals is an important facet of goal-directed behavior. The frontal cortex has been hypothesized to exert top-down inhibition of impulsive actions, but little is known about the nature of the neural signals responsible for such control. To address this question, we have developed a novel impulse control task in rats and begun to investigate the neural signals of frontal cortical areas, namely medial frontal cortex (MFC) and secondary motor cortex (M2), during the task.
In the impulse control task, subjects interact with a waiting port and a reward port. While waiting, two tones are generated, the first at a fixed short delay (0.4 s) and the second at a longer random delay (exponentially-distributed, min. 0.7 s, approx. 2s mean). Responses after the first tone garner a reward, but the amount is 3-4 times larger after the second tone. For a given set of reward amounts and delays, the time a rat is willing to wait varies randomly from trial to trial, approximating the distribution of second tone delays.
To characterize neural signals related to impulse control, we made single-unit recordings from MFC and M2 neurons, testing for signals that correlated on a trial-by-trial basis with waiting time. While approximately 20% of neurons in M2 (109/548 neurons) showed activity that correlated with and predicted waiting time, only 6.6% of MFC neurons (8/122 neurons) showed such a predictive activity. Waiting-predictive neurons in M2 showed a variety of time courses of activity. The most frequent one was a phasic signal locked to waiting-port entry, but other neurons showed ramping activity peaking at waiting-port exit.
To test whether the predictive activity in M2 is specific to waiting action or not, we compared the activity during two impulse control tasks which require different actions for waiting, nose-poke for one task and lever-press for the other. Most of the predictive signals are specific to one type of waiting, which suggests the action specificity of the predictive signal.
These results establish a task suitable for studying impulse control, reveal properties of single neuronal activity in frontal cortex during the task and suggest a role of M2 in planning timing of actions.

The striatum is an input structure of the basal ganglia implicated in several time-dependent functions including reinforcement learning, decision making, and interval timing. To determine whether striatal ensembles drive subjects' judgments of duration, we manipulated and recorded from striatal neurons in rats performing a duration categorization psychophysical task. We found that the dynamics of striatal neurons predicted duration judgments, and that simultaneously recorded ensembles could judge duration as well as the animal. Furthermore, striatal neurons were necessary for duration judgments, as muscimol infusions produced a specific impairment in animals' duration sensitivity. Lastly, we show that time as encoded by striatal populations ran faster or slower when rats judged a duration as longer or shorter, respectively. These results demonstrate that the speed with which striatal population state changes supports the fundamental ability of animals to judge the passage of time.

Ventral tegmental area (VTA) activity is critical for reward/reinforcement and is tightly modulated by the laterodorsal tegmentum (LDT). In utero exposure to glucocorticoids (iuGC) triggers prominent motivation deficits but nothing is known about the impact of this exposure in the LDT-VTA circuit. We show that iuGC-rats have long-lasting changes in cholinergic markers in the LDT, together with a decrease in LDT basal neuronal activity. Interestingly, upon LDT stimulation, iuGC animals present a decrease in the magnitude of excitation and an increase in VTA inhibition, as a result of a shift in the type of cells that respond to the stimulus. In agreement with LDT-VTA dysfunction, we show that iuGC animals present motivational deficits that are rescued by selective optogenetic activation of this pathway. Importantly, we also show that LDT-VTA optogenetic stimulation is reinforcing, and that iuGC animals are more susceptible to the reinforcing properties of LDT-VTA stimulation.

Descending modulation of nociceptive transmission depends on the release of noradrenaline at the spinal cord. The role of noradrenaline in the control of nociceptive transmission at the supraspinal pain control system remains understudied. As chronic pain is associated with enhanced descending facilitation of nociceptive transmission, we sought to determine the role of noradrenaline in pain facilitation from the brain during neuropathic pain. We determined the action of the noradrenergic input to the dorsal reticular nucleus (DRt), a unique pain facilitatory area, using the spared nerve injury model. Injections of the a1-adrenoreceptor agonist phenylephrine into the DRt induced hyperalgesia and allodynia, indicating that a1-adrenoreceptors enhance the facilitatory action of the nucleus. This led us to reduce noradrenaline release at the DRt using a viral vector derived from the Herpes Simplex virus type 1 (HSV-1) which carried the tyrosine hydroxylase (TH) transgene in antisense orientation. The reduction of noradrenaline release, confirmed by microdialysis experiments, induced a long-lasting attenuation of pain responses, which was reverted by the local administration of phenylephrine. The present study indicates that the noradrenergic modulation of a pronociceptive area at the supraspinal pain control system accounts for pain facilitation, through the activation of a1-adrenoreceptors. The study also shows that sustained effects on chronic pain can be achieved by decreasing the release of noradrenaline in a pain facilitatory centre of the brain using gene transfer.
