Abstract/Results: RESULTS:
Our study was originally concerned with full-length brain Cyclic Nucleotide Phosphodiesterase (CNP), a member of the 2H phosphodiesterase family, which is found abundantly in the myelin of oligodendrocytes in the central nervous system, and has been shown to be of major importance for axonal survival. As such, CNP is believed to be implicated in diseases such as multiple sclerosis and schizophrenia. The three-dimensional structure of the full-length protein is anticipated to yield new insights into the molecular mechanisms of the protein.
We have developed a protocol for expression and purification of the recombinant full-length protein, verified the integrity of the protein by mass spectrometry, and checked the fold and its stability by circular dichroism and fluorescence spectroscopy. Extensive crystallisation trials did not yield crystals suitable for X-ray diffraction so far. In selected trials, protein self-organisation was observed, probably indicating epitaxy-like formation of nanostructures on the glass plates.
Another family of brain proteins are the Visinin-like Proteins (VILIPs) that are neuronal calcium sensor proteins and an important factor for synaptic plasticity. They are involved in schizophrenia, neurodegenerative diseases such as Alzheimer’s and other disorders.
To obtain further insights into the molecular level mechanisms of VILIPs, homology models were generated and modelling techniques were used to elucidate the binding mechanisms of these proteins to specific membrane components (PIPs). We have developed the first three-dimensional model of VILIP:PIP complexesm, and propose a mechanism whereby recognition of specific PIP derivatives contributes to the targeting of VILIPs to subcellular locations.

The NCS protein Visinin-like Protein 1 (VILIP-1) transduces calcium signals in the brain and serves as an effector of the non-retinal receptor guanylyl cyclases (GCs) GC-A and GC-B, and nicotinic acetyl choline receptors (nAchR). Analysis of the quaternary structure of VILIP-1 in solution reveals the existence of monomeric and dimeric species, the relative contents of which are affected but not exclusively regulated by divalent metal ions and Redox conditions. Using small-angle X-ray scattering, we have investigated the low resolution structure of the calcium-bound VILIP-1 dimer under reducing conditions. Scattering profiles for samples with high monomeric and dimeric contents have been obtained. The dimerization interface involves residues from EF-hand regions EF3 and EF4.Using monolayer adsorption experiments, we show that myristoylated and unmyristoylated VILIP-1 can bind lipid membranes. The presence of calcium only marginally improves binding of the protein to the monolayer, suggesting that charged residues at the protein surface may play a role in the binding process.In the presence of calcium, VILIP-1 undergoes a conformational re-arrangement, exposing previously hidden surfaces for interaction with protein partners. We hypothesise a working model where dimeric VILIP-1 interacts with the membrane where it binds membrane-bound receptors in a calcium-dependent manner.

VILIP-1 belongs to the subfamily of Visinin-like proteins with its members VILIP-1, -2, -3, hippocalcin and neurocalcin d which have been implicated in the modulation of a variety of intracellular signaling cascades. VILIP-1 has recently been characterized in more detail and an involvement in cyclic nucleotide signaling has been demonstrated. Thus, similar to other NCS proteins such as GCAPs and neurocalcin d, VILIP-1 can influence the activtiy of guanylyl cyclase enzymes and the cGMP-signaling system of neurons in the nervous
system.

Among its activities the BIAL Foundation manages the BIAL Award, created in 1984, one of the most important awards in the Health field in Europe. The BIAL Award rewards both the basic and the clinical research distinguishing works of major impact in medical research.
The BIAL Foundation also assigns Scientific Research Scholarships for the study of neurophysiological and mental health in people, arousing the interest of researchers in the areas of Psychophysiology and Parapsychology.
To date the BIAL Foundation has supported 461 projects, more than 1000 researchers, with research groups in twenty-seven countries, resulting, until April 2013, in about 600 full papers, out of which 172 published in indexed international journals with an average impact factor of 3.6 and a substantial number of citations (1665).
Since 1996 the BIAL Foundation organizes the Symposia entitled "Behind and Beyond the Brain", a Forum that gathers well renowned neurosciences speakers and the BIAL Foundation Fellows which are spread around the world.
Classified as an institution of public utility, the BIAL Foundation includes among its patrons the Portuguese President, the Portuguese Universities Rectors' Council and the Portuguese Medical Association.

In 1994 the BIAL Foundation launched a programme of science research grants with the aim of encouraging the research into Man’s physical and mental processes, namely in fields still largely unexplored but which warrant further scientific analysis, as Psychophysiology and Parapsychology.
Since its launch, applications to the BIAL grants have been increasing. Up to now 461 projects have been supported, involving more than 1000 researchers from 27 countries.
The approved applications have benefited from grants in amounts comprised between €5,000 and €50, 000. The amount to be granted is fixed by the Scientific board according to the needs of each project.
The supported projects have originated, until April 2013, in about 600 full papers, 172 out of which were published in indexed international journals with an average impact factor of 3.6 and a substantial number of citations (1665).
Among the BIAL Foundation fellows is worth highlighting the presence of scientists from prestigious universities from the United States, United Kingdom, Australia, Russia, Germany, Japan, France, Canada, and many others.
The BIAL grants are promoted biannually.

OBJECTIVES:
Structure-function relationships of brain proteins need to be established in order to understand the molecular basis of brain disorders that affect organic health, as well as the psychological well-being.
METHODS:
Proteins have been prepared and purified, and subjected to structural investigation using spectroscopic and crystallographic methods. Molecular modelling has been used to assess protein-ligand binding.
RESULTS AND CONCLUSIONS:
Our study was originally concerned with full-length brain Cyclic Nucleotide Phosphodiesterase (CNP), a member of the 2H phosphodiesterase family, which is found abundantly in the myelin of oligodendrocytes in the central nervous system, and has been shown to be of major importance for axonal survival. As such, CNP is believed to be implicated in diseases such as multiple sclerosis and schizophrenia. The threedimensional structure of the full-length protein is anticipated to yield new insights into the molecular mechanisms of the protein. We have developed a protocol for expression and purification of the recombinant full-length protein, verified the integrity of the protein by mass spectrometry, and checked the fold and its stability by circular dichroism and fluorescence spectroscopy. Extensive crystallisation trials did not yield crystals suitable for X-ray diffraction so far. In selected trials, protein self-organisation was observed, probably indicating epitaxy-like formation of nanostructures on the glass plates. Another family of brain proteins are the Visinin-like Proteins (VILIPs) that are neuronal calcium sensor proteins and an important factor for synaptic plasticity. They are involved in schizophrenia, neurodegenerative diseases such as Alzheimer’s and other disorders.
To obtain further insights into the molecular level mechanisms of VILIPs, homology models were generated and modelling techniques were used to elucidate the binding mechanisms of these proteins to specific membrane components (PIPs). We have developed the first three-dimensional model of VILIP:PIP complexes, and propose a mechanism whereby recognition of specific PIP derivatives contributes to the targeting of VILIPs to subcellular locations.
