To date the BIAL Foundation has supported 461 projects, more than 1000 researchers, with research groups in twenty-seven countries, resulting, until April 2013, in about 600 full papers, out of which 172 published in indexed international journals with an average impact factor of 3.6 and a substantial number of citations (1665).
Since 1996 the BIAL Foundation organizes the Symposia entitled "Behind and Beyond the Brain", a Forum that gathers well renowned neurosciences speakers and the BIAL Foundation Fellows which are spread around the world.
Classified as an institution of public utility, the BIAL Foundation includes among its patrons the Portuguese President, the Portuguese Universities Rectors' Council and the Portuguese Medical Association.

Abstract/Results: ABSTRACT:
Williams-Beuren syndrome (WBS), a neurodevelopmental genetic disorder whose manifestations include visuospatial impairment, provides a unique model to link genetically determined loss of neural cell populations at different levels of the nervous system with neural circuits and visual behavior. Given that several of the genes deleted in WBS are also involved in eye development and the differentiation of retinal layers, we examined the retinal phenotype in WBS patients and its functional relation to global motion perception. We discovered a low-level visual phenotype characterized by decreased retinal thickness, abnormal optic disk concavity, and impaired visual responses in WBS patients compared with age-matched controls by using electrophysiology, confocal and coherence in vivo imaging with cellular resolution, and psychophysics. These mechanisms of impairment are related to the magnocellular pathway, which is involved in the detection of temporal changes in the visual scene. Low-level magnocellular performance did not predict high-level deficits in the integration of motion and 3D information at higher levels, thereby demonstrating independent mechanisms of dysfunction in WBS that will require remediation strategies different from those used in other visuospatial disorders. These findings challenge neurodevelopmental theories that explain cortical deficits based on low-level magnocellular impairment, such as regarding dyslexia.

OBJECTIVE:
Williams syndrome (WS) is characterized by visuospatial impairment often described as reflecting a local processing bias. However, these evidences are controversial, which may also be compounded by the large variability in performance that has been found across studies. To investigate local processing bias in WS and define whether this bias determines performance in global-local tasks.
PARTICIPANTS AND METHODS:
A preference task (match to sample similarity) and a performance task (requiring identification of local or global similarity) were conducted in 15 WS subjects using Navon hierarchical figures. Performance of WS group was compared to a chronological (n=15) and mental age-matched control group (n=15).
RESULTS:
The preference task demonstrated that 7 WS subjects showed a local bias, while 9 subjects performed like controls, exhibiting a global preference. Performance on the preference task was significantly correlated with education level and intelligence level. In the performance task WS subjects with a local processing bias made significantly more errors in the global condition than the WS subjects with a global preference.
CONCLUSIONS:
Our results suggest that the local bias is not homogeneous across WS subjects, thus reflecting the great variability described in other studies. By comparing WS subjects with a global bias to WS subjects with a local bias, we showed that the education level and the level of intelligence are important factors in defining the processing style. Additionally, the local processing preference found in some WS subjects seems to be related to their global processing deficit, both being commonly correlated with the education level.

A dorsal stream impairment has been described in WS. The dorsal stream is important for making allocentric judgments (viewer-independent) whereas egocentric spatial judgments (viewer-dependent) depend on the dorsal pathway. Here, we used two perceptual judgment tasks (computerized and ecological 3D tasks) to investigate egocentric and allocentric spatial perception in eighteen WS patients, as compared to Chronological (n=20) and mental (n=22) age-matched control groups. The WS group was impaired on the egocentric and allocentric task condition of the computerized task and the 3D task when compared with both control groups. Importantly, no selective impairment was found for perceiving egocentric representations, suggesting that dorsal stream function is not specifically impaired.

Why do some institutionalized children develop indiscriminate behavior (IB) while others do not? Considering children with Williams syndrome (WS) may provide an answer because IB has been observed routinely among individuals with this rare genetic neurodevelopmental disorder. By conceiving WS as a natural genetic model that mimics the indiscriminate phenotype and, more importantly, is associated with the deletion of genes in a specific region, we propose an integrative conceptual framework that underscores the dynamic developmental interplay between genes, endophenotypes, and environment. In this article, we consider the etiology of IB among institutionalized children, which emphasizes environmental factors, followed by the effect of such behavior on WS children's hypersociability, which highlights the crosstalk between genes and neuropsychological features in programming their distinctive social-emotional and behavioral phenotype. We propose new hypotheses regarding the etiopathogeny of IBs in institutionalized children, particularly the prediction of specific Gene × Environment interactions.

Abstract/Results: ABSTRACT:
Williams syndrome (WS) is a clinical model of dorsal stream vulnerability and impaired visual integration. However, little is still known about the neurophysiological correlates of perceptual integration in this condition. We have used a 3D structure-from-motion (SFM) integrative task to characterize the neuronal underpinnings of 3D perception in WS and to probe whether gamma oscillatory patterns reflect changed holistic perception. Coherent faces were parametrically modulated in 3D depth (three different depth levels) to vary levels of stimulus ambiguity. We have found that the electrophysiological (EEG/ERP) correlates of such holistic percepts were distinct across groups. Independent component analysis demonstrated the presence of a novel component with a late positivity around 200 ms that was absent in controls. Source localization analysis of ERP signals showed a posterior occipital shift in WS and reduced parietal dorsal stream sources. Interestingly, low gamma-band oscillations (20-40 Hz) induced by this 3D perceptual integration task were significantly stronger and sustained during the stimulus presentation in WS whereas high gamma-band oscillations (60-90 Hz) were reduced in this clinical model of impaired visual coherence, as compared to controls. These observations suggest that dorsal stream processing of 3D SFM stimuli has distinct neural correlates in WS and different cognitive strategies are employed by these patients to reach visual coherence. Importantly, we found evidence for the presence of different sub-bands (20-40 Hz/60-90 Hz) within the gamma range which can be dissociated concerning the respective role on the coherent percept formation, both in typical and atypical development.

Object and depth perception from motion cues involves the recruitment of visual dorsal stream brain areas. In 3-D structure-from-motion (SFM) perception, motion and depth information are first extracted in this visual stream to allow object categorization, which is in turn mediated by the ventral visual stream. Such interplay justifies the use of SFM paradigms to understand dorsal-ventral integration of visual information. The nature of such processing is particularly interesting to be investigated in a neurological model of cognitive dissociation between dorsal (impaired) and ventral stream (relatively preserved) processing, Williams syndrome (WS). In the current fMRI study, we assessed dorsal versus ventral stream processing by using a performance-matched 3-D SFM object categorization task. We found evidence for substantial reorganization of the dorsal stream in WS as assessed by whole-brain ANOVA random effects analysis, with subtle differences in ventral activation. Dorsal reorganization was expressed by larger medial recruitment in WS (cuneus, precuneus, and retrosplenial cortex) in contrast with controls, which showed the expected dorsolateral pattern (caudal intraparietal sulcus and lateral occipital cortex). In summary, we found a substantial reorganization of dorsal stream regions in WS in response to simple visual categories and 3-D SFM perception, with less affected ventral stream. Our results corroborate the existence of a medial dorsal pathway that provides the substrate for information rerouting and reorganization in the presence of lateral dorsal stream vulnerability. This interpretation is consistent with recent findings suggesting parallel routing of information in medial and lateral parts of dorsal stream.
