Abstract/Results: RESULTS:
The studies included 3 groups of subjects: control group (C - 15 subjects), schizophrenic patients (S - 35 subjects), volunteers, exposed to 6-day dry immersion (DI - 12 subjects). All participants had right hand and right eye preference. Subjects performed visually-guided saccades and antisaccades. EEG was recorded from 19 standard sites. Mean amplitude of averaged potentials was assessed at 600 ms before the stimulus and at 600 to 100 ms before saccade onset.
The saccade characteristics did not differ in groups C and DI. Patients exhibited in the antisaccade task delays in the performance of correct saccades (p<0,05) and larger number of directional errors (p<0,001).
In control subjects the target stimulus was preceded by a vertex predominantly bilateral slow negative potential shift; left- and right-side saccades were preceded by PSN, that was larger at the midline and left sites, with parietal maximum before visually-guided saccades, and with frontal and parietal maximums before antisaccades. The PSN amplitude declined significantly in the DI and Sch groups as compared with the C group. However, in immersed subjects the foci of negativity shifted to the right hemisphere so that the PSN amplitude decreased sharply in the left and increased in the right hemisphere, with significant decrease in Fz, F3, Pz and P3. Significant decline of the PSN amplitude was the most characteristic in schizophrenic patients in the sagittal frontal and central regions (Fz and ?z).
The PSN shifts observed after exposure to simulated weightlessness and in schizophrenic patients could be caused by the alterations of sensory inputs’ activities in weightlessness and fine frontal structural disorder of the brain in case of schizophrenia.

Contingent negative variation (CNV) topography, hemispheric asymmetry and time-course were investigated in healthy subjects and non-medicated paranoid schizophrenic patients in two antisaccade paradigms with the short (800-1000 ms) and long (1200-1400 ms) durations of the fixation period. EEG and electrooculogram (EOG) were recorded. Saccade characteristics and mean amplitudes of slow cortical potentials time-locked to peripheral target were analyzed in 23 healthy volunteers and 19 schizophrenic patients. Compared to healthy control subjects, schizophrenic patients had significantly slower antisaccades and committed significantly more erroneous saccades in the both antisaccade tasks. The prolongation of the fixation period resulted in noticeable decrease of error percent in patients group. The analysis of CNV time-course has revealed two distinct stages in both groups. The early CNV stage was represented by a negative wave with the maximal amplitude over midline fronto-central area, and the late stage was characterized by increased CNV amplitude at the midline and left parietal electrode sites. In healthy subjects the simultaneous activation of frontal and parietal areas was observed in the paradigm with the shorter fixation interval; the increase of the fixation period produced consecutive activation of these areas. Schizophrenic patients' CNV amplitude was generally smaller than that of healthy subjects. The most pronounced between-group differences of the negative shift amplitude were revealed at frontal electrode sites during the early CNV stage in both modifications of the antisaccade task. The deficit of frontal activation revealed in patients at the early stage of antisaccade preparatory set in both antisaccadic paradigms may be related to pathogenesis of paranoid schizophrenia.

Slow cortical potentials preceding visually guided saccades in schizophrenics

The parameters of saccades and presaccadic slow potentials were studied in right-handed men with a dominant right eye, including 19 schizophrenics and 12 healthy subjects. For visual stimulation, three light-emitting diodes were used, which were located in the center of the visual field (the central fixation stimulus) and 10° to the right and left of it (peripheral stimuli). Two stimulation protocols were used: with a simultaneous switching off of the central fixation stimulus and switching on of the peripheral stimuli (test 1) and with an interstimulus gap of 200 ms (test 2). According to the latency, saccades were divided into anticipatory, express, and regular. Slow EEG potentials preceding regular saccades were analyzed. It was found that the proportion of anticipatory saccades is considerably higher than the normal value in schizophrenia. The analysis of the presaccadic potentials demonstrated a significant decrease in the amplitude of negative potentials in the vertex region at early stages of presaccadic preparation and its increase in the occipital region at late stages. Test 2 in the patients demonstrated an increase in the positivity focus in the frontal region of the right hemisphere. It was assumed that the alterations found in schizophrenia result from the deficit of frontal cortical fields.

Antisaccade task performance and mean amplitudes of slow cortical potentials (contingent negative variation—CNV) were investigated in 19 healthy volunteers, 16 schizophrenic patients (SP), and 12 patients with stereotyped form of paraphilia (PP). Compared with healthy subjects, schizophrenic and paraphilic patients committed significantly more erroneous saccades. The clear between-group CNV differences were observed during the early CNV stage that is associated with cognitive aspects of preparatory set. In SP, as compared to controls, the significant decline of CNV amplitude was found at frontal-central area. PP have demonstrated the lack of CNV over central and parietal regions, but their CNV amplitudes in frontal area did not differ from values of control group. Thus, two distinct types of CNV abnormalities have been found. The SP results have been interpreted as support for frontal dysfunction in schizophrenia. The disconnection between prefrontal cortex, sensorimotor cortex, and related subcortical structures is hypothesized in paraphilia group.
