Final report - Life-Span Changes in Electrophysiological Patterns Associated with Temporal Discrimination

Abstract/Results: RESULTS:
The present project aimed at investigating neural correlates of time processing in the life-span. To this aim three lines of research were carried out.
The first line concerned the development of new methodological tools for the analysis of EEG signal.
A novel channel reactivity based (CRB) method has been created for individual alpha frequency (IAF) computation. The CRB method is based on quantitative indexes and criteria and relies on task-specific alpha reactivity patterns rather than on the presence of peaks in the EEG spectrum. In order to improve time-frequency analyses of EEG data, we applied a wavelet-based methodology using a continuous wavelet transform with a complex Morlet as mother function. Furthermore, event-related potentials (ERPs) were measured using a single-trial approach to estimate the intra individual variability (IIV) of P300 parameters (latency and amplitude).
The second line identified cognitive and neural mechanisms underlying time processing in young and older adults.
The overall findings were very promising. By testing healthy subjects, we could clarify cognitive processes engaged in time processing, especially related to memory. Furthermore, by means of the EEG/ERP methodology we could elucidate the role of frontal and parietal brain regions involved in such processing, and their development and decline with age.
The third line examined cognitive performance and electrophysiological patterns associated with timing and related cognitive processes in pathological populations, such as minimal hepatic encephalopathy, Parkinson’s disease, preterm newborns, and ADHD.
Both behavioral and ERP data in patients supported the hypothesis that the processing of time in the range of hundred milliseconds to few seconds is impaired in such clinical populations. Results might further have implications in the diagnostic and rehabilitation protocols.

The individual alpha frequency (IAF) is one of the most common tools used to study the variability of EEG rhythms among subjects. Several approaches have been proposed in the literature for IAF determination, including the popular peak frequency (PF) method, the extended band (EB) method, and the transition frequency (TF) method. However, literature techniques for IAF determination are over-reliant on the presence of peaks in the EEG spectrum and are based on qualitative criteria that require visual inspection of every individual EEG spectrum, a task that can be time consuming and difficult to reproduce. In this paper a novel channel reactivity based (CRB) method is proposed for IAF computation. The CRB method is based on quantitative indexes and criteria and relies on task-specific alpha reactivity patterns rather than on the presence of peaks in the EEG spectrum. Application of the technique to EEG signals recorded from 19 subjects during a cognitive task demonstrates the effectiveness of the CRB method and its capability to overcome the limits of PF, EB, and TF approaches.

The Syntactic and Semantic Processing of Mass and Count Nouns: An ERP Study

The present study addressed the question of whether count and mass nouns are differentially processed in the brain. In two different ERP (Event-Related Potentials) tasks we explored the semantic and syntactic levels of such distinction. Mass and count nouns typically differ in concreteness, hence the effect of this important variable was factorially examined in each task. Thus the stimuli presented were: count concrete, count abstract, mass concrete or mass abstract. The first experiment (concrete/abstract semantic judgment task) involved the interaction between the N400 concreteness effect and the Mass/ Count condition, revealing a substantial effect between mass and count nouns at the semantic level. The second experiment (sentence syntactic violation task) showed a Mass/Count distinction on left anterior negativity (LAN) and on P600 components, confirming the difference at the syntactic level. This study suggests that the brain differentiates between count and mass nouns not only at the syntactic level but also at the semantic level. Implications for our understanding of the brain mechanisms underlying the Mass/Count distinction are discussed.

In 1994 the BIAL Foundation launched a programme of science research grants with the aim of encouraging the research into Man’s physical and mental processes, namely in fields still largely unexplored but which warrant further scientific analysis, as Psychophysiology and Parapsychology.
Since its launch, applications to the BIAL grants have been increasing. Up to now 461 projects have been supported, involving more than 1000 researchers from 27 countries.
The approved applications have benefited from grants in amounts comprised between €5,000 and €50, 000. The amount to be granted is fixed by the Scientific board according to the needs of each project.
The supported projects have originated, until April 2013, in about 600 full papers, 172 out of which were published in indexed international journals with an average impact factor of 3.6 and a substantial number of citations (1665).
Among the BIAL Foundation fellows is worth highlighting the presence of scientists from prestigious universities from the United States, United Kingdom, Australia, Russia, Germany, Japan, France, Canada, and many others.
The BIAL grants are promoted biannually.

Abstract/Results: ABSTRACT:
Prospective memory (PM) is the ability to remember to accomplish an action when a particular event occurs (i.e., event-based PM), or at a specific time (i.e., time-based PM) while performing an ongoing activity. Strategic Monitoring is one of the basic cognitive functions supporting PM tasks, and involves two mechanisms: a retrieval mode, which consists of maintaining active the intention in memory; and target checking, engaged for verifying the presence of the PM cue in the environment. The present study is aimed at providing the first evidence of event-related potentials (ERPs) associated with time-based PM, and at examining differences and commonalities in the ERPs related to Strategic Monitoring mechanisms between event- and time-based PM tasks.
The addition of an event-based or a time-based PM task to an ongoing activity led to a similar sustained positive modulation of the ERPs in the ongoing trials, mainly expressed over prefrontal and frontal regions. This modulation might index the retrieval mode mechanism, similarly engaged in the two PM tasks. On the other hand, two further ERP modulations were shown specifically in an event-based PM task. An increased positivity was shown at 400–600 ms post-stimulus over occipital and parietal regions, and might be related to target checking. Moreover, an early modulation at 130–180 ms post-stimulus seems to reflect the recruitment of attentional resources for being ready to respond to the event-based PM cue. This latter modulation suggests the existence of a third mechanism specific for the event-based PM; that is, the “readiness mode”.

Abstract/Results: ABSTRACT:
The aim of the present study was to analyze and compare time monitoring and event monitoring processes in two prospective memory (PM) tasks. For this purpose a time-based PM task and an event-based PM task were designed, which included the same ongoing task and the same frequency and timing of PM targets. The analysis of reaction times in the ongoing task in a baseline block (only ongoing) and the PM-block (ongoing + PM) was conducted.
Mean reaction times (RTs) of the ongoing task in the time-based condition were significantly lower than RTs in the baseline block, suggesting that the PM task minimally interfere with the ongoing. Furthermore, ongoing RTs were inversely correlated with the frequency of clock checks, meaning individuals who monitored the clock less frequently rely more heavily on their "internal clock"; such mechanism subtracts cognitive resources to the ongoing task.
Overall, mean RTs of the ongoing task in the event-based condition did not differ from RTs in the baseline block and were higher than the time-based task. This result might reflect a continuous monitoring of the target. A detailed analysis of ongoing RTs highlights specific mechanisms of monitoring in time- and event-based PM tasks.

OBJECTIVE:
Intra-individual variability (IIV) of response reaction times (RTs) and psychomotor slowing were proposed as markers of brain dysfunction in patients with minimal hepatic encephalopathy (MHE), a subclinical disorder of the central nervous system frequently detectable in patients with liver cirrhosis. However, behavioral measures alone do not enable investigations into the neural correlates of these phenomena. The aim of this study was to investigate the electrophysiological correlates of psychomotor slowing and increased IIV of RTs in patients with MHE.
METHODS:
Event-related potentials (ERPs), evoked by a stimulus–response (S–R) conflict task, were recorded from a sample of patients with liver cirrhosis, with and without MHE, and a group of healthy controls. A recently presented Bayesian approach was used to estimate single-trial P300 parameters.
RESULTS:
Patients with MHE, with both psychomotor slowing and higher IIV of RTs, showed higher P300 latency jittering and lower single-trial P300 amplitude compared to healthy controls. In healthy controls, distribution analysis revealed that single-trial P300 latency increased and amplitude decreased as RTs became longer; however, in patients with MHE the linkage between P300 and RTs was weaker or even absent.
CONCLUSIONS:
These findings suggest that in patients with MHE, the loss of the relationship between P300 parameters and RTs is related to both higher IIV of RTs and psychomotor slowing.
SIGNIFICANCE:
This study highlights the utility of investigating the relationship between single-trial ERPs parameters along with RT distributions to explore brain functioning in normal or pathological conditions.

Age-related differences in the neural correlates of remembering time-based intentions

The present study used event-related potentials (ERPs) to explore the effect of age on the neural correlates of monitoring processes involved in time-based prospective memory. In both younger and older adults, the addition of a time-based prospective memory task to an ongoing task led to a sustained ERP activity broadly distributed over the scalp. Older adults, however, did not exhibit the slow wave activity observed in younger adults over prefrontal regions, which is considered to be associated with retrieval mode. This finding indicates that age-related decline in intention maintenance might be one source of the impaired prospective memory performance displayed by older adults. An 'anterior shift' in scalp distribution of the P3 was observed in older adults, and was related to lower levels of accuracy in prospective memory performance. This relationship suggests that possible factors responsible for age-related decline in prospective memory performance include the decreased efficiency of executive/frontal functions as well as the reduced amount of resources available for the prospective memory task.

The time course of temporal discrimination: An ERP study

Abstract/Results: ABSTRACT:
OBJECTIVE - The question of how temporal information is processed by the brain is still a matter of debate. This study aimed to elucidate the brain electrical activity associated with a visual temporal discrimination task.
METHODS - For this purpose, 44 participants were required to compare pairs of sequentially presented time intervals: a fixed standard interval (1000ms), and an equal-to-standard, longer (1200ms) or shorter (800ms) comparison interval. Behavioural data and event-related potentials (ERPs) were analyzed.
RESULTS - Long intervals were more rapidly identified than short intervals. The amplitude of the contingent negative variation (CNV) found at frontocentral sites before the end of the comparison interval was significantly affected by the difference between its duration and the standard one. The amplitude and the scalp distribution of ERPs registered after the offset of the comparison interval were linearly modulated by its absolute duration.
CONCLUSIONS - ERP components associated with the offset of the comparison intervals clarified the involvement of working memory processes and different brain structures in temporal discrimination.
SIGNIFICANCE - This study further improves our understanding of the cognitive processes and neural substrates underlying temporal discrimination in healthy subjects and lays the ground for the investigation of clinical samples with time processing deficits.

Abstract/Results: ABSTRACT:
EEG signals recorded by surface electrodes placed on the scalp can be thought as nonstationary
stochastic processes in both time and space, especially in response to external stimuli.
Cognitive tasks, in particular, are reflected by changes in EEG dynamics concerning both rhythms
energy and connectivity across different brain regions. In the frequency-domain, EEG analysis is
complicated and time-frequency methodologies are needed. The Wavelet Transform, in particular,
represents a powerful tool for analysing, within a time-frequency embedding, the EEG. In this study
we applied a wavelet-based methodology to extract quantitative time-frequency parameters from EEG
signals recorded during a time discrimination task in 12 subjects. We used a continous wavelet
transform with a complex Morlet as mother function. In order to improve the time-frequency resolution
and to make it satisfactory, each of the four standard EEG rhythms (i.e. theta, alpha, beta, gamma) was
studied with Morlet wavelet parameters tuned ad hoc on the basis of both the width of the specific
frequency band and the particular type of activity under examination. The numerical values of the
estimated time-frequency indexes were then compared, evidencing statistically significant differences
in the brain response between experimental conditions.

To date, neural correlates associated with time-base prospective memory (TB-PM) tasks have been scarcely investigated. The present study explored electrophysiological correlates of a TB-PM task. Fourteen participants were administered with two experimental conditions: an ongoing task alone (baseline) and an ongoing + PM task (TB-PM). Electrical potentials locked to the ongoing stimuli were compared between the two conditions. The results showed higher and more frontal positive amplitude in 180-300 ms time window in the TB-PM condition compared to the baseline. Furthermore, a significant amplitude increase in 400-600 ms time window emerged over centro-parietal sites. The amplitude modulations associated to TB-PM instructions in anterior sites are supposed to reflect higher recruitment of attentional resources engaged in time monitoring. The higher amplitude observed in parietal sites mirrors EEG findings of event-based PM tasks, which are related to the maintenance of intention.

Background - Cortical auditory evoked potentials (CAEPs) may serve as an early index of developmental problems in the
auditory cortex.
Aim - To determine the effects of neonatal risk factors on maturational changes of CAEPs components.
Methods: Auditory processing was investigated before discharge in forty-two NICU admitted premature infants (mean
gestational age-GA 30 weeks, range 23-34). Recordings consisted of the average of 150 responses to 1000 Hz pure tones (inter-stimulus interval 1250 1850 ms) recorded at frontocentral locations. CAEP components were correlated with GA, gender, bronco-pulmonary dysplasia (BPD), retinopathy of prematurity (ROP), intra-ventricular haemorrhage (IVH),
intrauterine growth restriction (IUGR), resuscitation at birth, patent ductus arteriosus (PDA), infections, use of antenatal
steroids.
Results - CAEP components were significantly correlated with GA (p = 0.001), BPD (p = .002), ROP (p = .02), IVH (p = .019)
and resuscitation (p = .045); moreover ROP and BPD were significantly higher in infants born at GA< 29 weeks and the
occurrence of ROP or BPD was the only significant finding in a multivariate analysis.
Conclusions - Cortical auditory processing appears to be influenced by neonatal clinical data associated with fetal
inflammatory response, complicating extremely preterm birth. These findings points to the importance of early risk factors on brain development.

Abstract/Results: ABSTRACT:
We propose a Bayesian method to extract single-trial event related potentials (ERPs). The method is formulated in two stages. In the first stage, each of the N raw sweeps is processed by an individual "optimal" filter, where the 2nd order a priori statistical information on the background EEG and on the unknown ERP is, respectively, estimated from pre-stimulus data and obtained through the multiple integration of a white noise process model which is identifiable from post-stimulus data thanks to a smoothing criterion. Then, a mean ERP is determined as the weighted average of the filtered sweeps, where each weight is inversely proportional to the expected value of the norm of the correspondent filter error. In the second stage, single-sweep estimation is dealt with within the same framework, by using the average ERP estimated in the previous stage as a priori expected response. The method is successfully tested on simulated data and then employed on real data with the aim of investigating the variability of the P300 component during a cognitive visual task. A comparison with other literature methods is also performed. Results encourage further use of the proposed method to investigate if and how diseases, e.g., cirrhosis, are associated to differences in P300 variability.

Monitoring mechanisms in time- and event-based prospective memory

Abstract/Results: ABSTRACT:
The aim of the present study was to analyze and compare time monitoring and event monitoring processes in two prospective memory (PM) tasks. For this purpose a time-based PM task and an event-based PM task were designed, which included the same ongoing task and the same frequency and timing of PM targets. The analysis of reaction times in the ongoing task in a baseline block (only ongoing) and the PM-block (ongoing + PM) was conducted.
Mean reaction times (RTs) of the ongoing task in the time-based condition were significantly lower than RTs in the baseline block, suggesting that the PM task minimally interfere with the ongoing. Furthermore, ongoing RTs were inversely correlated with the frequency of clock checks, meaning individuals who monitored the clock less frequently rely more heavily on their "internal clock"; such mechanism subtracts cognitive resources to the ongoing task.
Overall, mean RTs of the ongoing task in the event-based condition did not differ from RTs in the baseline block and were higher than the time-based task. This result might reflect a continuous monitoring of the target. A detailed analysis of ongoing RTs highlights specific mechanisms of monitoring in time- and event-based PM tasks.

Several approaches, based on different assumptions and with various degree of theoretical sophistication and implementation complexity, have been developed for extracting the single-trial response of event related potentials (ERPs). In many of these methods, one of the major challenges is the exploitation of a priori knowledge. In this paper, we present a new method where the 2nd order statistical information on the background EEG and on the unknown ERP, necessary for the optimal filtering of each sweep in a Bayesian estimation framework, is, respectively, estimated from pre-stimulus data and obtained through a multiple integration of a white noise process model. The latter model is flexible and simple enough to be easily identifiable from the post-stimulus data thanks to a smoothing criterion. A mean ERP is determined as the weighted average of the filtered sweeps, where each weight is inversely proportional to the expected value of the norm of the correspondent filter error, a quantity determinable thanks to the employment of the Bayesian approach. Then, single-sweep estimation is dealt with within the same framework. The method is tested on simulated data and compared with a recently proposed estimation method based on radial-basis function (RBF) neural networks. Then, the method is also employed on real data with the aim of investigating the variability of the P300 component in cirrhotic vs normal subjects undertaken to a cognitive visual task.

Objectives: The aim of the study was to compare the spectral values of EEG sleep states in preterm
infants born at different gestational age who were at the same 35 weeks of post-conceptional age
(PCA).
Methods: One-hour monopolar EEG was recorded before discharge in twenty-one NICU admitted
premature infants (mean gestational age-GA 30 weeks, range 27–35) during active (AS) and quiet
sleep (QS) from electrodes Fp1, Fp2, Fz, T3, T4, C3, Cz, C4, O1 and O2. Fast Fourier transform
algorithm was applied for power spectral analysis of the EEG. The existence of changes between
sleep states and gestational age in the power of delta, theta, alpha, beta, low gamma and gamma
bands was tested.
Results: The results of the spectral measures show a power increase in the low frequency bands
(delta and theta) positively correlated with gestational age in active sleep, which can be mainly
traced on fronto-central electrodes (p<.002). The power in the higher frequency bands remains
homogeneous between subjects. No significant differences are found in quiet sleep.
Conclusions: At the same post-conceptional age, differences in brain electrical activity are highlight
between premature infants depending on their gestational age and sleep state. The differences
are present in the low frequency bands, the most prominent activity in the immature brain of the
preterm neonate.
Discussion: Spectral EEG power, an index of local neural synchrony, is considered the strongest
indicator of maturation after 36 PCA (Scher et al., 1995). The lack of changes in the higher
frequencies is compatible with the assumption that generation of these oscillations typically
requires well-formed structural intra-cortical inhibitory circuitry which are not yet mature at this
developmental phase (Kostovic and Judas, 2002). Interesting, the detected differences in our study
are present only in active sleep, the sleep state in that occurs endogenous neurosensory
stimulation, fundamental process for cortical development and cognitive processing (Graven,
2006, Bisiacchi et al 2009). These results may reflect alterations in brain function as a results of
conditions leading to extreme premature birth and/or of prolonged adaption to extra-uterine life.
These findings show that differences are evident even before term age and points to the
importance of early identification of risk factors on brain development.

How we compute time is not fully understood. Questions include whether an automatic brain mechanism is engaged in temporally regular environmental structure in order to anticipate events, and whether this can be dissociated from task-related processes, including response preparation, selection and execution. To investigate these issues, a passive temporal oddball task requiring neither time-based motor response nor explicit decision was specifically designed and delivered to participants during high-density, event-related potentials recording. Participants were presented with pairs of audiovisual stimuli (S1 and S2) interspersed with an Inter-Stimulus Interval (ISI) that was manipulated according to an oddball probabilistic distribution. In the standard condition (70% of trials), the ISI lasted 1,500 ms, while in the two alternative, deviant conditions (15% each), it lasted 2,500 and 3,000 ms. The passive over-exposition to the standard ISI drove participants to automatically and progressively create an implicit temporal expectation of S2 onset, reflected by the time course of the Contingent Negative Variation response, which always peaked in correspondence to the point of S2 maximum expectation and afterwards inverted in polarity towards the baseline. Brain source analysis of S1- and ISI-related ERP activity revealed activation of sensorial cortical areas and the supplementary motor area (SMA), respectively. In particular, since the SMA time course synchronised with standard ISI, we suggest that this area is the major cortical generator of the temporal CNV reflecting an automatic, action-independent mechanism underlying temporal expectancy.
