ABSTRACT:
Over the past years increasing amount of attention has been given to signaling lipids as well as to its modulating enzymes, such as phospholipases. Specifically, phospholipase D (PLD), that converts phosphatidylcholine to phosphatidic acid (PA), has been shown to exhibit a role in neurological development and physiology. Several studies have been associating PLD1 and PLD2, the two main mammalian PLD isozymes, to neurological processes, including neurotransmitter release, dendritic branching, cognition, and brain development. Also, the hippocampus has been suggested as one of the brain regions showing the highest PLD activity and neurodegenerative conditions such as Alzheimer’s disease associated pathways have been shown to be modulated by PLD signaling. Thus, the aim of this project was to better understand the potential role of PLD in hippocampal function in adult mice upon Pld1 or Pld2 genetic ablation.
We performed a hippocampal related behavioral characterization of these animals, a structural analysis regarding dendritic morphology, electrophysiology neuronal plasticity evaluation and protein and lipidomic biochemical detailed analysis.
Our behavioral data, specifically considering motor and exploratory activity, anxiety and memory, showed that PLD2 knockout mice behavior is not altered when compared to their wild type littermates. Although most of this was also observed in the animals lacking PLD1, the results indicate an object recognition-dependent short-term memory deficit. Regarding hippocampal dendritic arborization, the ablation of either PLD1 or PLD2 led to significant alterations in dendritic morphology, although with a different impact of each isozyme in the dorsal and ventral hippocampus. Concerning the electrophysiological plasticity analysis we observed a deficit in long-term depression in the dorsal hippocampus of PLD1 knock-out mice. Finally our biochemical analysis revealed a significant decrease in SNAP-25 protein levels and total levels of PA levels in PLD1 knock-out mice while no major changes were observed upon PLD2 genetic ablation.
Our results suggest that the separate ablation of PLD1 and PLD2 leads to different effects in the dorsal and ventral hippocampus, with a more significant effect upon PLD1 ablation.

Lipids are a major constituent of the brain and lipid metabolic defects have been implicated in neurodegenerative diseases.
Phospholipase D (PLD) is a lipid-modulating enzyme and, in mammals, there are two main PLD isozymes - PLD1 and PLD2, which generate phosphatidic acid from phosphatidylcholine, affecting signaling lipid and membrane trafficking processes. The hippocampus has been suggested as one of the brain regions showing the highest PLD activity and at the pathological level, PLD was shown to play a role in Alzheimer’s disease (AD) pathogenesis. Also, it was recently shown that the hippocampus presents different electrophysiological properties with differential functions along its longitudinal axis. While the dorsal hippocampus (DH) is predominantly involved in spatial navigation and episodic memory, the ventral hippocampus (VH) is more involved in emotional related behaviors. Since synaptic hippocampal plasticity is fundamental in learning processes and since it was shown to be impaired in the setting of AD, we propose to study the impact of PLD2 ablation in adult hippocampal synaptic functioning along its dorso-ventral axis. Dorsal hippocampus (DH) and ventral hippocampus (VH) slices were prepared from PLD2 wild-type (WT) and PLD2 knock-out (KO) and used for extracellular recordings. In the input-output relationship and paired-pulse facilitation ratio, PLD2 ablation had no significant impact. In long-term depression (LTD) inducibility, while we found no changes upon PLD2 ablation
in the DH, in the VH we observed an increased excitability in the early LTD induction phase in PLD2 KO animals. In summary, we show that PLD2 ablation does not affect hippocampal plasticity properties in the DH and that it leads to earlyphase LTD induction deficit in the VH. Future studies addressing the molecular associated pathways in early-phase LTD will shed light in PLD2-dependent mechanisms.

Lipids are major constituents of the brain largely implicated in physiological and pathological processes. The hippocampus is a complex brain structure involved in learning, memory and emotional responses, and its functioning is also affected in various disorders. Despite conserved intrinsic circuitry, behavioral and anatomical studies suggest the existence of a structural and functional gradient along the hippocampal longitudinal axis. Here, we used an unbiased mass spectrometry approach to characterize the lipid composition of distinct hippocampal subregions. In addition, we evaluated the susceptibility of each area to lipid modulation by corticosterone (CORT), an important mediator of the effects of stress. We confirmed a great similarity between hippocampal subregions relatively to other brain areas. Moreover, we observed a continuous molecular gradient along the longitudinal axis of the hippocampus, with the dorsal and ventral extremities differing significantly from each other, particularly in the relative abundance of sphingolipids and phospholipids. Also, whereas chronic CORT exposure led to remodeling of triacylglycerol and phosphatidylinositol species in both hippocampal poles, our study suggests that the ventral hippocampus is more sensitive to CORT-induced changes, with regional modulation of ceramide, dihydrosphingomyelin and phosphatidic acid. Thus, our results confirm a multipartite molecular view of dorsal-ventral hippocampal axis and emphasize lipid metabolites as candidate effectors of glucocorticoid signaling, mediating regional susceptibility to neurological disorders associated with stress.

The BIAL Foundation also assigns Scientific Research Scholarships for the study of neurophysiological and mental health in people, arousing the interest of researchers in the areas of Psychophysiology and Parapsychology.
To date the BIAL Foundation has supported 461 projects, more than 1000 researchers, with research groups in twenty-seven countries, resulting, until April 2013, in about 600 full papers, out of which 172 published in indexed international journals with an average impact factor of 3.6 and a substantial number of citations (1665).
Since 1996 the BIAL Foundation organizes the Symposia entitled "Behind and Beyond the Brain", a Forum that gathers well renowned neurosciences speakers and the BIAL Foundation Fellows which are spread around the world.
Classified as an institution of public utility, the BIAL Foundation includes among its patrons the Portuguese President, the Portuguese Universities Rectors' Council and the Portuguese Medical Association.

In 1994 the BIAL Foundation launched a programme of science research grants with the aim of encouraging the research into Man’s physical and mental processes, namely in fields still largely unexplored but which warrant further scientific analysis, as Psychophysiology and Parapsychology.
Since its launch, applications to the BIAL grants have been increasing. Up to now 461 projects have been supported, involving more than 1000 researchers from 27 countries.
The approved applications have benefited from grants in amounts comprised between €5,000 and €50, 000. The amount to be granted is fixed by the Scientific board according to the needs of each project.
The supported projects have originated, until April 2013, in about 600 full papers, 172 out of which were published in indexed international journals with an average impact factor of 3.6 and a substantial number of citations (1665).
Among the BIAL Foundation fellows is worth highlighting the presence of scientists from prestigious universities from the United States, United Kingdom, Australia, Russia, Germany, Japan, France, Canada, and many others.
The BIAL grants are promoted biannually.

Abstract/Results: ABSTRACT:
Phosphatidic acid (PA) is a signaling lipid involved in the modulation of synaptic structure and functioning. Based on previous work showing a decreasing PA gradient along the longitudinal axis of the rodent hippocampus, we asked whether the dorsal hippocampus (DH) and the ventral hippocampus (VH) are differentially affected by PA modulation. Here, we show that phospholipase D1 (PLD1) is a major hippocampal PA source, compared to PLD2, and that PLD1 ablation affects predominantly the lipidome of the DH. Moreover, Pld1 knockout (KO) mice show specific deficits in novel object recognition and social interaction and disruption in the DH-VH dendritic arborization differentiation in CA1/CA3 pyramidal neurons. Also, Pld1 KO animals present reduced long-term depression (LTD) induction and reduced GluN2A and SNAP-25 protein levels in the DH. Overall, we observe that PLD1-derived PA reduction leads to differential lipid signatures along the longitudinal hippocampal axis, predominantly affecting DH organization and functioning.
