Abstract/Results: ABSTRACT:
Neurons in higher cortical areas, such as the prefrontal cortex, are often tuned to a variety of sensory and motor variables, and are therefore said to display mixed selectivity. This complexity of single neuron responses can obscure what information these areas represent and how it is represented. In this project we developed and demonstrated advantages of a new dimensionality reduction technique, demixed principal component analysis (dPCA), that decomposes population activity into a few components. In addition to systematically capturing the majority of the variance of the data, dPCA also exposes the dependence of the neural representation on task parameters such as stimuli, decisions, or rewards. To illustrate our method we reanalyzed population data from four datasets comprising different species, different cortical areas and different experimental tasks. In each case, dPCA provides a concise way of visualizing the data that summarizes the task-dependent features of the population response in a
single figure.
This project also involved some additional work on dimensionality reduction of electrophysiological datasets, including data from auditory cortex and from input areas to dopaminergic system in rats.

Abstract/Results: ABSTRACT:
Dopamine neurons encode the difference between actual and predicted reward, or reward prediction error (RPE). Although many models have been proposed to account for this computation, it has been difficult to test these models experimentally. Here we established an awake electrophysiological recording system, combined with rabies virus and optogenetic cell-type identification, to characterize the firing patterns of monosynaptic inputs to dopamine neurons while mice performed classical conditioning tasks. We found that each variable required to compute RPE, including actual and predicted reward, was distributed in input neurons in multiple brain areas. Further, many input neurons across brain areas signaled combinations of these variables. These results demonstrate that even simple arithmetic computations such as RPE are not localized in specific brain areas but, rather, distributed across multiple nodes in a brain-wide network. Our systematic method to examine both activity and connectivity revealed unexpected redundancy for a simple computation in the brain.

The BIAL Foundation also assigns Scientific Research Scholarships for the study of neurophysiological and mental health in people, arousing the interest of researchers in the areas of Psychophysiology and Parapsychology.
To date the BIAL Foundation has supported 461 projects, more than 1000 researchers, with research groups in twenty-seven countries, resulting, until April 2013, in about 600 full papers, out of which 172 published in indexed international journals with an average impact factor of 3.6 and a substantial number of citations (1665).
Since 1996 the BIAL Foundation organizes the Symposia entitled "Behind and Beyond the Brain", a Forum that gathers well renowned neurosciences speakers and the BIAL Foundation Fellows which are spread around the world.
Classified as an institution of public utility, the BIAL Foundation includes among its patrons the Portuguese President, the Portuguese Universities Rectors' Council and the Portuguese Medical Association.

Constraints on the accuracy of auditory discriminations in rats

Does variability of sensory neurons constrain the accuracy of perception? This question has vexed researchers for decades. Answering it requires making sure that an animal’s behavioral accuracy in a sensory discrimination task is exclusively limited by sensory factors, something which to the best of our knowledge has not yet been shown in rodents. Here we first provide evidence that this is the case in a 2AFC inter-aural level difference (ILD) discrimination task in rats. We show that, in this task, behavioral accuracy cannot be improved by manipulations of motivation, and that accuracy is unaffected by past trials in most subjects. Given that performance is apparently Limited by ‘sensory noise’, we then studied the structure of neural variability in auditory cortex. As a first step in this direction, we have quantified neural variability in the absence of behavior. Specifically, we simultaneously recorded the evoked responses of large neural populations (N  100) from the auditory cortex of rats under urethane anesthesia, using the same set of stimuli as in the ILD discrimination task. We made three key observations: (1) although approximately half of the neurons are ILD-selective, single-cell neurometric functions systematically underperform compared to behavior, except for < 1% of the neurons. (2) Population-level neurometric functions can match behavioral performance, but show a strong brain-state dependence, with performance increasing with the level of desynchronization. Finally, we show that this brain-state dependence arises from a reorganization of where signal and noise reside in the high-dimensional space of population firing rates. Our results establish a behavioral paradigm in rats for the investigation of a long-standing question in neuroscience, and constitute one of the first empirical investigations of the relationship between neural variability and behavioral accuracy at the population level.

Neurons in higher cortical areas, such as the prefrontal cortex, are often tuned to a variety of sensory and motor variables, and are therefore said to display mixed selectivity. This complexity of single neuron responses can obscure what information these areas represent and how it is represented. Here we present a dimensionality reduction technique, demixed principal component analysis (dPCA), that decomposes population activity into a few components. In addition to systematically capturing the majority of the variance of the data, dPCA also exposes the neural representation in terms of task parameters such as stimuli, decisions, or rewards. To illustrate our method we reanalyze population data from four datasets comprising different species, different cortical areas and different experimental tasks. In each case, dPCA provides a concise way of visualizing the data that summarizes all the relevant features of the population response in a single figure. In addition, dPCA highlights several important and seemingly universal features of the population activity in prefrontal cortex; e.g. that most of the neural activity is not related to any of the controlled task parameters, such as stimuli or decision; or that there are no well-separated functional clusters of cells.

The accuracy of the neural code depends on the relative embedding of signal and noise in the activity of neural populations. Despite a wealth of theoretical work on population codes, there are few empirical characterizations of the high-dimensional signal and noise subspaces. We studied the geometry of population codes in the rat auditory cortex across brain states along the activation-inactivation continuum, using sounds varying in difference and mean level across the ears. As the cortex becomes more activated, single-hemisphere populations go from preferring contralateral loud sounds to a symmetric preference across lateralizations and intensities, gain-modulation effectively disappears, and the signal and noise subspaces become approximately orthogonal to each other and to the direction corresponding to global activity modulations. Level-invariant decoding of sound lateralization also becomes possible in the active state. Our results provide an empirical foundation for the geometry and state-dependence of cortical population codes.
