Abstract/Results: ABSTRACT:
Alzheimer’s disease (AD) is the most common form of dementia worldwide, characterized by progressive memory impairment, behavioral changes, and, ultimately, loss of consciousness and death. Recently, microRNA (miRNA) dysfunction has been associated with increased production and impaired clearance of amyloid-ß (Aß) peptides, whose accumulation is one of the most well-known pathophysiological markers of this disease. In this study, we identified several miRNAs capable of targeting key proteins of the amyloidogenic pathway. The expression of one of these miRNAs, miR-31, previously found to be decreased in AD patients, was able to simultaneously reduce the levels of APP and Bace1 mRNA in the hippocampus of 17-month-old AD triple-transgenic (3xTg-AD) female mice, leading to a significant improvement of memory deficits and a reduction in anxiety and cognitive inflexibility. In addition, lentiviral-mediated miR-31 expression significantly ameliorated AD neuropathology in this model, drastically reducing Aß deposition in both the hippocampus and subiculum. Furthermore, the increase of miR-31 levels was enough to reduce the accumulation of glutamate vesicles in the hippocampus to levels found in non-transgenic age-matched animals. Overall, our results suggest that miR-31-mediated modulation of APP and BACE1 can become a therapeutic option in the treatment of AD.

Social hierarchies are present in most mammalian species. In nature, hierarchies offer a tradeoff between reduction of in-group fighting between males, at the expense of an asymmetric sharing of resources. Early life experiences and stress are known to influence the rank an individual attains in adulthood, but the associated cellular and synaptic alterations are poorly understood. Using a maternal separation protocol, we show that care-deprived mice display a long-lasting submissive phenotype, increased social recognition, and enhanced explorative behavior. These alterations are consistent with an adaptation that favors exploration rather than confrontation within a group setting. At the neuronal level, these animals display dendritic atrophy and enhanced inhibitory synaptic inputs in medial prefrontal cortex (mPFC) neurons. To determine what could underlie this synaptic modification, we first assessed global gene expression changes via RNAseq, and next focused on a smaller subset of putatively altered synaptic receptors that could explain the changes in synaptic inhibition. Using different cohorts of maternally deprived mice, we validated a significant increase in the expression of Npy1r, a receptor known to play a role in maternal care, anxiety, foraging, and regulation of group behavior. Using electrophysiological recordings in adult mice while blocking NPY1R signaling, we determined that this receptor plays a key role in enhancing GABAergic currents in mice that experience maternal deprivation. Taken together, our work highlights the potential of regulating NPY1R in social anxiety disorders and the alterations induced in brain circuitry as a consequence of early life stress and adversity.

BACKGROUND
Exposure to early-life stress (ELS), such as that caused by maternal separation can induce maladaptive behaviors and increase the vulnerability to neurological disorders later in life. In addition to having a direct impact in neuronal function, stress can also trigger neuroinflammatory events that impact microglia activity.
AIMS
With this work, we aim to elucidate what is the acute and long-term impact of ELS exposure and how it interferes with key microglia functions which are crucial for correct circuit wiring during the post-natal period.
METHOD
Towards this purpose, we have used a paradigm of maternal separation and maternal stress (MSUS), that allows to mimic early life adversity in the form of maternal neglect. We have investigated consequences of ELS to social behavior, as well as its impact on microglia phenotype. We have focused our evaluation in the medial pre-frontal cortex (mPFC), a brain region implicated in social interaction and impulse control that is often implicated in neuropsychiatric disorders.
RESULTS
We have observed that exposure to MSUS causes changes in social interaction, in the form of a reduction in ultravocalizations, as early as P5. In addition, we observed changes in social preference and an increase in submissive behaviors in adolescent and adult male mice, but no significant behavior changes in females. Exposure to ELS also leads to sex-dependent morphological and gene expression changes that are more profound in male microglia and that originate an hyperramified phenotype in adolescent mice. Male MSUS mice also present an increase in the number of PV+ inhibitory neurons in the mPFC, that is not observed in females and that leads to an increase in spontaneous IPSCs in this region.
CONCLUSIONS
Taken together, ours results point towards a higher susceptibility of male mice to the negative impacts of ELS exposure. This feature may be directly connected with the different and delayed response of male microglia to this type of stress.

Childhood exposure to stress in forms of abuse or neglect is common worldwide. Reports have demonstrated that early life stress is associated with overall volume loss, as well as with reduced neuronal density in several brain regions. Furthermore, an increasingly number of clinical studies have demonstrated a clear relationship between stressful events in the early-life period and neuropsychiatric/neurodevelopmental disorders such as depression, autism spectrum disorders, schizophrenia, psychosis and post-traumatic stress disorders.The mechanisms underlying this relationship are not clear, but microglia cells have been suggested as mediators of the interaction between stress and alterations in neuronal circuits, which underly these pathologies. These cells, that until recently were believed to remain in a resting state until challenged, are now known to participate in several neurodevelopmental processes essential for correct formation of the brain network. Being a part of the immune system, microglia cells are very sensitive to environmental cues and pro-inflammatory stimulus such as stress in early-life stages. Moreover, alterations in these cells during the critical periods of development lead to impairments in microglia-mediated processes and these impairments have already been correlated with neuronal circuit alterations. However, most of these mechanisms have only been described in male animals, with little information available concerning the impact of early-life stress in the sexual dysmorphism that is present during brain development under physiological conditions.The present study aimed to investigate the impact of early-life stress in microglia morphology, density and function in the medial prefrontal cortex, a brain region known to be particularly relevant in processes such as complex cognitive function, decision making and social cognition, as well as its impact in social and anxiety-like behaviors during adolescence, focusing in the differences between genders. Our results revealed that early-life stress causes alterations in social behavior and triggers depressive and risk-taking behaviors in male adolescent animals but not in females. In addition, exposure to this type of stress led a general activation of microglia cells and to opposite changes in microglia density and morphology in both genders, highlighting the sexual dysmorphic characteristics of these cells. Although further investigation in this subject is still warranted, we hypothesize that the differences observed in microglia phenotype can contribute to the increase resilience observed in females following stress in early-life, a result that might help clarify the different incidence of several neuropsychiatric disorders between genders.

Despite great advances in neuroscience research, knowledge on the mechanisms behind the origin of neuropsychiatric disorders, as well as possible therapies, are still lacking. Recently, the neuroimmune system has been studied in the context of these diseases, with emphasis given to microglia, due to their role in brain development. Microglia are known to regulate the pool of neuronal precursors and aid in neuronal circuit formation, as well as having a role in brain homeostasis in adulthood. Microglia constantly scan the brain for potential threats, undergoing immune activation in response to varied stimuli, then triggering inflammatory processes. In the context of neuropsychiatric diseases, activated microglia have been found in the brain of patients. Moreover, microglia dysfunction in critical periods of brain maturation has been hypothesized to contribute to circuitry alterations underlying these disorders.Microglia are heterogenous throughout different brain regions. Due to particularities of microglia in the cerebellum, including an enhanced clearance phenotype and activation priming, and the involvement of this brain region in regulating motor and social behaviors, the main goal of this thesis was to understand if challenging microglia function in critical periods of cerebellar maturation contributes to circuitry and behavioral alterations. In this work, it was showed that the maturation of cerebellar microglia is heavily hindered by IL-4, a Th2 anti-inflammatory cytokine considered as neuroprotective in neurodegeneration. IL-4 injection in mice not only affected microglia morphology, density and distribution in the cerebellum, but also induced alterations in Purkinje cell connectivity, ultimately leading to long-lasting behavioral abnormalities, such as motor coordination deficits, enhanced mobility and altered sociability. This work highlights the vulnerability of cerebellar maturation to environmental stimuli during the second postnatal week.

Prolonged exposure to stressful conditions in early life imparts changes to physiology and health and may trigger or exacerbate neuropsychiatric disorders. Overt dysfunction in sociability is also a hallmark of disorders such as autism spectrum disorder (ASD) and schizophrenia. Likewise, abnormal social interaction in otherwise healthy individuals is itself a trigger for mental health disorders. In addition, for most social animals, the relative position an individual occupies in its social hierarchy strongly modulates psychosocial stress. Early life stress (ELS) induced by maternal separation is acknowledged to have long-term effects on animal behavior and is considered one of the most potent, naturally-occurring stressors. In humans, childhood adversity, neglect or mistreatment is known to increase the risk for psychiatric disorders, including depression and anxiety. In rodent models of maternal separation, behavioral deficits are observed in emotional behavior and stress responses. At the same time, neurological dysfunctions have been identified in both humans and mice exposed to ELS. However, the precise neuronal substrates and mechanisms that traduce ELS into altered neuronal activity and subsequent social dysfunction in adulthood are poorly understood. While the role of stress impacting adult animals and promoting changes to social behavior and social hierarchies is well described, the influence of ELS on adult social hierarchical rank and social behaviors remains largely unknown. Our results uncovered that ELS induces a dysfunction in social hierarchy and social interactions in mice. We observed that ELS mice display a lower social rank when compared to control animals and are more easily defeated in dominant-subordinate competitions. ELS also led to long-lasting functional alterations in the medial prefrontal cortex (mPFC), as evidenced by an increased c-fos expression in response to social dominance stimuli. Additionally, neuronal atrophy and increased inhibitory synaptic strength were observed in mPFC neurons from adult mice that experience ELS. In terms of gene expression in the mPFC, we found that the mRNA of Npy1r was upregulated in ELS mice and also in wild-type subordinate animals, suggesting a role for this gene in behaviors regulating social dominance. Together, our data suggests that ELS leads to long-lasting alterations in gene expression and to altered mPFC activity, inducing a subordinate behavior within a social hierarchy. Lastly, we proposed Npy1r signaling as a key modulator of social rank and a potential therapeutic target for social dysfunctions.

Among all major organs, the brain is one of the most susceptible to the inexorable effects of aging. Throughout the last decades, several studies in human cohorts and animal models have revealed a plethora of age-related changes in the brain, including reduced neurogenesis, oxidative damage, mitochondrial dysfunction and cell senescence. As the main immune effectors and first responders of the nervous tissue, microglia are at the center of these events. These cells experience irrevocable changes as a result from cumulative exposure to environmental triggers, such as stress, infection and metabolic dysregulation. The age-related immunosenescent phenotype acquired by microglia is characterized by profound modifications in their transcriptomic profile, secretome, morphology and phagocytic activity, which compromise both their housekeeping and defensive functions. As a result, aged microglia are no longer capable of establishing effective immune responses and sustaining normal synaptic activity, directly contributing to age-associated cognitive decline and neurodegeneration. This review discusses how lifestyle and environmental factors drive microglia dysfunction at the molecular and functional level, also highlighting possible interventions to reverse aging-associated damage to the nervous and immune systems.

Along the years the BIAL Foundation has developed an important relationship with the scientific community, first in Portugal and after worldwide. Today it is an institution of reference which aims to stimulate new researches that may help people, promote more health and contribute to new milestones to gain access to knowledge.
Among its activities the BIAL Foundation manages the BIAL Award, created in 1984, one of the most important awards in the Health field in Europe. The BIAL Award rewards both the basic and the clinical research distinguishing works of major impact in medical research.
The BIAL Foundation also assigns Scientific Research Scholarships for the study of neurophysiological and mental health in people, arousing the interest of researchers in the areas of Psychophysiology and Parapsychology.
To date the BIAL Foundation has supported 461 projects, more than 1000 researchers, with research groups in twenty-seven countries, resulting, until April 2013, in about 600 full papers, out of which 172 published in indexed international journals with an average impact factor of 3.6 and a substantial number of citations (1665).
Since 1996 the BIAL Foundation organizes the Symposia entitled "Behind and Beyond the Brain", a Forum that gathers well renowned neurosciences speakers and the BIAL Foundation Fellows which are spread around the world.
Classified as an institution of public utility, the BIAL Foundation includes among its patrons the Portuguese President, the Portuguese Universities Rectors' Council and the Portuguese Medical Association.

In 1994 the BIAL Foundation launched a programme of science research grants with the aim of encouraging the research into Man’s physical and mental processes, namely in fields still largely unexplored but which warrant further scientific analysis, as Psychophysiology and Parapsychology.
Since its launch, applications to the BIAL grants have been increasing. Up to now 461 projects have been supported, involving more than 1000 researchers from 27 countries.
The approved applications have benefited from grants in amounts comprised between €5,000 and €50, 000. The amount to be granted is fixed by the Scientific board according to the needs of each project.
The supported projects have originated, until April 2013, in about 600 full papers, 172 out of which were published in indexed international journals with an average impact factor of 3.6 and a substantial number of citations (1665).
Among the BIAL Foundation fellows is worth highlighting the presence of scientists from prestigious universities from the United States, United Kingdom, Australia, Russia, Germany, Japan, France, Canada, and many others.
The BIAL grants are promoted biannually.

Microglia are tissue-resident macrophages responsible for the surveillance, neuronal support, and immune defense of the brain parenchyma. Recently, the role played by microglia in the formation and function of neuronal circuits has garnered substantial attention. During development, microglia have been shown to engulf neuronal precursors and participate in pruning mechanisms while, in the mature brain, they influence synaptic signaling, provide trophic support and shape synaptic plasticity. Recently, studies have unveiled different microglial characteristics associated with specific brain regions. This emerging view suggests that the maturation and function of distinct neuronal circuits may be potentially associated with the molecular identity microglia adopts across the brain. Here, we review and summarize the known role of these cells in the thalamus, hippocampus, cortex, and cerebellum. We focus on in vivo studies to highlight the characteristics of microglia that may be important in the remodeling of these neuronal circuits and in relation to neurodevelopmental and neuropsychiatric disorders.
