Abstract/Results: ABSTRACT:
Influential theories suggest that a defensive behavioral inhibition system (BIS) supports the inhibition of action tendencies when facing potential threats. However, little is known about threat-related inhibitory mechanisms in humans and their relations to inter-individual differences in BIS sensitivity. To address this issue, we used paired-pulse TMS to investigate early human motor cortex (M1) responses to social signals of potential threats, like another’s fearful body posture. In two experiments, participants observed pictures of fearful and happy postures, and neutral postures that were either dynamic (in Exp1) or static (in Exp2). To test suppression of M1 excitatory activity, we assessed intracortical facilitation (ICF) in an early phase of threat monitoring by administering TMS pulses at 100–125 ms from picture onset. We investigated the motor representation of hand and arm muscles that are differentially involved in flexion, extension, and abduction. As a control, we also assessed corticospinal excitability and short intracortical inhibition. In both experiments, and independently of the muscle, watching fearful bodies suppressed ICF relative to watching happy and non-emotional (dynamic or static) body expressions. Remarkably, greater fear-related ICF suppression was found in participants who scored higher on a self-report questionnaire assessing BIS sensitivity. These findings suggest that observing fearful body language activates a defensive suppression of M1 excitatory activity that is influenced by the personality disposition to experience fear and anxiety when facing potential threats. This BIS-related motor suppression may have the functional role of transiently suppressing action tendencies to promote threat monitoring and, ultimately, survival.

The primary motor cortex (M1) is highly influenced by premotor/motor areas both within and across hemispheres. Dual site transcranial magnetic stimulation (dsTMS) has revealed interhemispheric interactions mainly at early latencies. Here, we used dsTMS to systematically investigate long-latency causal interactions between right-hemisphere motor areas and the left M1 (lM1). We stimulated lM1 using a suprathreshold test stimulus (TS) to elicit motor-evoked potentials (MEPs) in the right hand. Either a suprathreshold or a subthreshold conditioning stimulus (CS) was applied over the right M1 (rM1), the right ventral premotor cortex (rPMv), the right dorsal premotor cortex (rPMd) or the supplementary motor area (SMA) prior to the TS at various CS-TS inter-stimulus intervals (ISIs: 40-150 ms). The CS strongly affected lM1 excitability depending on ISI, CS site and intensity. Inhibitory effects were observed independently of CS intensity when conditioning PMv, rM1 and SMA at a 40-ms ISI, with larger effects after PMv conditioning. Inhibition was observed with suprathreshold PMv and rM1 conditioning at a 150-ms ISI, while site-specific, intensity-dependent facilitation was detected at an 80-ms ISI. Thus, long-latency interhemispheric interactions, likely reflecting indirect cortico-cortical/cortico-subcortical pathways, cannot be reduced to nonspecific activation across motor structures. Instead, they reflect intensity-dependent, connection- and time-specific mechanisms.

Mind-wandering, the mind’s capacity to stray from external events and generate task-unrelated thought, has been associated with activity in the brain default network. To date, little is understood about the contribution of individual nodes of this network to mind-wandering. Here, we investigated the role of medial prefrontal cortex (mPFC) in mind-wandering, by perturbing this region with transcranial direct current stimulation (tDCS). Young healthy participants performed a choice reaction time task both before and after receiving cathodal tDCS over mPFC, and had their thoughts periodically sampled. We found that tDCS over mPFC - but not occipital or sham tDCS - decreased the propensity to mind-wander. The tDCS-induced reduction in mind-wandering occurred in men, but not in women, and was accompanied by a change in the content of task-unrelated though, which became more related to other people (as opposed to the self) following tDCS. These findings indicate that mPFC is crucial for mind-wandering, possibly by helping construction of self-relevant scenarios capable to divert attention inward, away from perceptual reality. Gender-related differences in tDCS-induced changes suggest that mPFC controls mind-wandering differently in men and women, which may depend on differences in the structural and functional organization of distributed brain networks governing mind-wandering, including mPFC.

Abstract/Results: ABSTRACT:
Functional imaging studies suggest that accurate understanding of others' emotional feelings (i.e., empathic accuracy, EA) recruits high-order visual, sensorimotor and mentalizing brain networks. However, the behavioral relevance of these findings is unclear. To fill in this gap, we used repetitive transcranial magnetic stimulation (rTMS) to interfere with the right superior temporal sulcus (STS), inferior frontal gyrus (IFG) and temporoparietal junction (TPJ) during an EA task requiring participants to infer the enjoyment felt by a social target while smiling/laughing. Relative to a baseline condition (sham rTMS), active rTMS of STS, IFG and TPJ (but not of a control site) disrupted the efficiency of EA task performance, mainly by lowering task accuracy; rTMS of IFG and TPJ also slowed down response speeds. Importantly, the effects of rTMS on EA task efficiency were predicted by baseline EA performance, with high-performers showing a performance decrease when the TPJ was targeted, and low-performers showing a performance decrease when the STS or the IFG was targeted. The double dissociation in the effect of rTMS between low- and high- performers suggests distinct roles of STS, IFG and TPJ in efficient understanding of the enjoyment felt by others. These findings provide causal evidence of distinct visual, sensorimotor and cognitive routes to EA and suggest that individual differences in EA are underpinned by differential recruitment of these routes.

Humans show exquisite abilities to perform versatile finger movements. The inferior frontal cortex (IFC) plays a pivotal role in the visual control of such movements through connections with other sensorimotor regions. Yet, the dynamics of IFC effective connectivity during action execution are still poorly understood. Using single-pulse TMS and simultaneous EEG recording (i.e., TMS-EEG coregistration), we stimulated the left posterior IFC at rest and during a visuomotor task. We recorded TMS-evoked potentials (TEPs) to assess action-related changes in IFC connectivity and localized their sources using sLORETA. We found two key time windows at ~60 and ~80 msec after IFC stimulation in which TEPs were modulated by task conditions in remote electrodes. In the first time window (~60 msec), action-related changes in TEP amplitudes were observed over frontal and temporo-parietal electrodes, reflecting increased IFC connectivity with fronto-parietal motor areas and decreased IFC connectivity with visual occipito-temporal areas. In the second time window (~80 msec), action-related TEP increases were observed in frontal, temporal and parietal regions partially overlapping with the default-mode network. No similar effects were observed when TMS was administered over a non-motor control area (the left posterior superior temporal sulcus, STS). These findings highlight dynamic changes in IFC connectivity with motor, sensory and default-mode networks. They suggest sequential stages of task-related changes in IFC connectivity possibly related to controlling and sensing actions and inhibiting default-mode brain activity during motor performance.

Skillful goal-directed manual actions such as grasping and manipulating objects are supported by a large sensorimotor network. Within this network, the ventral premotor cortex (PMv) transforms visual information about objects into motor commands that are conveyed to the primary motor cortex (M1), allowing fine control of finger movements. However, it is unknown whether transcranial magnetic stimulation (TMS) of this PMv-to-M1 hierarchical pathway improves action performance. To fill in this gap, here, we used cortico-cortical paired associative stimulation (ccPAS) with the aim of manipulating synaptic efficacy in the human PMv-to-M1 pathway. We found that repeatedly pairing TMS of pre-and post-synaptic nodes of the PMv-to-M1 pathway (i.e., PMv-to-M1 ccPAS) increased motor excitability and enhanced performance on the 9-Hole Peg Test (9-HPT), which taps into PMv-M1 functioning. These effects were specific to the ccPAS protocol consistent with the direction of the PMv-to-M1 hierarchy, as no effects were observed when reversing the order of the paired TMS pulses (i.e., following a M1-to-PMv ccPAS) or when administering sham ccPAS. Additionally, the effect of PMv-to-M1 ccPAS appeared functionally specific, as no behavioral enhancement was observed in a visuomotor control task. We therefore provide novel causal evidence that the PMv-to-M1 pathway, which is instrumental to object-oriented hand actions, is sensitive to TMS manipulations of associative plasticity. Our study highlights the causal role of the PMv-to-M1 pathway in controlling skillful object-oriented hand actions and suggests that ccPAS might be a useful tool for investigating the functional relevance of directional connectivity in humans. These findings may have implications for designing novel therapeutic strategies based on the manipulation of associative plasticity in cortico-cortical networks.

Witnessing another person’s suffering elicits vicarious brain activity in areas that are active when we ourselves are in pain. Whether this activity influences prosocial behavior remains the subject of debate. Here participants witnessed a confederate express pain through a reaction of the swatted hand or through a facial expression, and could decide to reduce that pain by donating money. Participants donate more money on trials in which the confederate expressed more pain. Electroencephalography shows that activity of the somatosensory cortex I (SI) hand region explains variance in donation. Transcranial magnetic stimulation (TMS) shows that altering this activity interferes with the pain–donation coupling only when pain is expressed by the hand. High-definition transcranial direct current stimulation (HD-tDCS) shows that altering SI activity also interferes with pain perception. These experiments show that vicarious somatosensory activations contribute to prosocial decision-making and suggest that they do so by helping to transform observed reactions of affected body-parts into accurate perceptions of pain that are necessary for decision-making.

ABSTRACT:
Influential theories suggest that humans predict others’ upcoming actions by using their own motor system as an internal forward model. However, evidence that the motor system is causally essential for predicting others’ actions is meager. Using transcranial direct current stimulation (tDCS), we tested the role of the inferior frontal cortex (IFC), in action prediction (AP). We devised a novel AP task where participants observed the initial phases of right-hand reaching-to-grasp actions and had to predict their outcome (i.e., the goal/object to be grasped). We found that suppression by cathodal (inhibitory) tDCS of the left IFC, but not the left superior temporal sulcus or the right IFC, selectively impaired performance on the AP task, but not on a difficulty-matched control task. Remarkably, anodal (excitatory) tDCS of the left IFC brought about a selective improvement in the AP task. These findings indicate that the left IFC is necessary for predicting the outcomes of observed human right-hand actions. Crucially, our study shows for the first time that down- and up-regulating excitability within the motor system can hinder and enhance AP abilities, respectively. These findings support predictive coding theories of action perception and have implications for enhancement of AP abilities.

Primary motor cortex crucial for action prediction: A tDCS study

Abstract/Results: ABSTRACT
The neural network underlying action observation - i.e., the action observation network - forms an anticipatory representation of observed actions. Although correlational studies suggest that the motor cortex (M1) might be involved in this anticipatory coding, it is unclear whether M1 is also causally essential for making accurate predictions about observed actions. To test the functional relevance of M1 to action prediction, we used offline monopolar transcranial direct current stimulation (tDCS). In four tDCS groups of healthy participants, we administered 15 min of anodal or cathodal constant currents of 1 or 2 mA over the left M1 before participants performed two tasks requiring them to make predictions about the outcomes of reaching-grasping human actions (Action Prediction - AP) or non-human movements (Non-human Prediction - NP). In each group, participants received sham and active tDCS in two separate sessions. We found that 2 mA cathodal tDCS (c-tDCS2mA) selectively impaired accuracy in the AP task, but not in the NP task. No change in performance was found following anodal or 1-mA tDCS protocols. Additionally, no change was found following c-tDCS2mA administered over a control site. These findings show task-, polarity-, intensity- and site-specific disruption of AP abilities following c-tDCS2mA over M1. Thus, our study establishes specific tDCS parameters for effective M1 stimulation in AP and highlights the functional relevance of the motor system to making accurate predictions about the outcomes of human actions.

Witnessing another person’s suffering elicits vicarious brain activity in areas that are active when we ourselves are in pain. Whether this activity influences prosocial behavior remains the subject of debate. Here participants witnessed a confederate express pain through a reaction of the swatted hand or through a facial expression, and could decide to reduce that pain by donating money. Participants donate more money on trials in which the confederate expressed more pain. Electroencephalography shows that activity of the somatosensory cortex I (SI) hand region explains variance in donation. Transcranial magnetic stimulation (TMS) shows that altering this activity interferes with the pain–donation coupling only when pain is expressed by the hand. High-definition transcranial direct current stimulation (HD-tDCS) shows that altering SI activity also interferes with pain perception. These experiments show that vicarious somatosensory activations contribute to prosocial decision-making and suggest that they do so by helping to transform observed reactions of affected body-parts into accurate perceptions of pain that are necessary for decision-making.

ABSTRACT
BACKGROUND
Accurately recognizing emotional expressions is critical for social life. Perceiving others’ emotional expressions activates a complex visual network including the primary visual cortex (V1) and higher-order regions such as the superior temporal sulcus (STS) which is known to encode biological motion. Yet, a fundamental and unanswered question is how STS and V1 interact to give rise to accurate emotion recognition.
AIMS
The ambitious aim of the present project is to shed new lights on the way the human visual system makes humans aware of emotional stimuli and as, ultimate goal, by improving its functioning, enhance visual emotion discrimination abilities.
METHOD
Using a novel Transcranial Magnetic Stimulation protocol, we transiently enhanced synaptic efficiency in the reentrant connections from STS to V1 and we tested facial emotion discrimination performances and possible neurophysiological changes (i.e., modulations in the visual evoked potentials recorded by electroencephalography).
RESULTS
We found that boosting the STS-V1 connectivity enhances emotion discrimination abilities as well as modulates the amplitude of early visual components involved in emotional face perception or in cognitive performance.
CONCLUSIONS
We provide novel causal evidence that STS-to-V1 back-projections, are malleable and functionally relevant to emotion recognition. These findings have implications for theoretical models of visual perception and awareness and for the rehabilitation of visual deficits.

Abstract/Results: ABSTRACT:
Growing neurophysiological evidence points to a role of alpha oscillations in divergent thinking (DT). In particular, studies have shown a consistent EEG alpha synchronization during performance on the Alternative Uses Task (AUT), a well-established DT task. However, there is a need for investigating the brain dynamics underlying the production of a sequence of multiple, alternative ideas at the AUT and their relationship with idea originality. In twenty young adults, we investigated changes in alpha power during performance on a structured version of the AUT, requiring to ideate four alternative uses for conventional objects in distinct and sequentially balanced time periods. Data analysis followed a three-step approach, including behaviour aspects, physiology aspects, and their mutual relationship. At the behavioural level, we observed a typical serial order effect during DT production, with an increase of originality associated with an increase in ideational time and a decrease in response percentage over the four responses. This pattern was paralleled by a shift from alpha desynchronization to alpha synchronization across production of the four alternative ideas. Remarkably, alpha power changes were able to explain response originality, with a differential role of alpha power over different sensor sites. In particular, alpha synchronization over frontal, central, and temporal sites was able to predict the generation of original ideas in the first phases of the DT process, whereas alpha synchronization over centro-parietal sites persistently predicted response originality during the entire DT production. Moreover, a bilateral hemispheric effect in frontal sites and a left-lateralized effect in central, temporal, and parietal sensor sites emerged as predictors of the increase in response originality. These findings highlight the temporal dynamics of DT production across the generation of alternative ideas and support a partially distinct functional role of specific cortical areas during DT.

The BIAL Foundation was created in 1994 by Laboratórios BIAL in conjunction with the Council of Rectors of Portuguese Universities. BIAL’s Foundation mission is to foster the scientific study of Man from both the physical and spiritual perspectives.
Along the years the BIAL Foundation has developed an important relationship with the scientific community, first in Portugal and after worldwide. Today it is an institution of reference which aims to stimulate new researches that may help people, promote more health and contribute to new milestones to gain access to knowledge.
Among its activities the BIAL Foundation manages the BIAL Award, created in 1984, one of the most important awards in the Health field in Europe. The BIAL Award rewards both the basic and the clinical research distinguishing works of major impact in medical research.
The BIAL Foundation also assigns Scientific Research Scholarships for the study of neurophysiological and mental health in people, arousing the interest of researchers in the areas of Psychophysiology and Parapsychology.
To date the BIAL Foundation has supported 461 projects, more than 1000 researchers, with research groups in twenty-seven countries, resulting, until April 2013, in about 600 full papers, out of which 172 published in indexed international journals with an average impact factor of 3.6 and a substantial number of citations (1665).
Since 1996 the BIAL Foundation organizes the Symposia entitled "Behind and Beyond the Brain", a Forum that gathers well renowned neurosciences speakers and the BIAL Foundation Fellows which are spread around the world.
Classified as an institution of public utility, the BIAL Foundation includes among its patrons the Portuguese President, the Portuguese Universities Rectors' Council and the Portuguese Medical Association.

In 1994 the BIAL Foundation launched a programme of science research grants with the aim of encouraging the research into Man’s physical and mental processes, namely in fields still largely unexplored but which warrant further scientific analysis, as Psychophysiology and Parapsychology.
Since its launch, applications to the BIAL grants have been increasing. Up to now 461 projects have been supported, involving more than 1000 researchers from 27 countries.
The approved applications have benefited from grants in amounts comprised between €5,000 and €50, 000. The amount to be granted is fixed by the Scientific board according to the needs of each project.
The supported projects have originated, until April 2013, in about 600 full papers, 172 out of which were published in indexed international journals with an average impact factor of 3.6 and a substantial number of citations (1665).
Among the BIAL Foundation fellows is worth highlighting the presence of scientists from prestigious universities from the United States, United Kingdom, Australia, Russia, Germany, Japan, France, Canada, and many others.
The BIAL grants are promoted biannually.

The presence of a network of areas in the parietal and premotor cortices, which are active both during action execution and observation, suggests that we might understand the actions of other people by activating those motor programs for making similar actions. Although neurophysiological and imaging studies show an involvement of the somatosensory cortex (SI) during action observation and execution, it is unclear whether SI is essential for understanding the somatosensory aspects of observed actions. To address this issue, we used off-line transcranial magnetic continuous theta-burst stimulation (cTBS) just before a weight judgment task. Participants observed the right hand of an actor lifting a box and estimated its relative weight. In counterbalanced sessions, we delivered sham and active cTBS over the hand region of the left SI and, to test anatomical specificity, over the left motor cortex (Ml) and the left superior parietal lobule (SPL). Active cTBS over SI, but not over Ml or SPL, impaired task performance relative to sham cTBS. Moreover, active cTBS delivered over SI just before participants were asked to evaluate the weight of a bouncing ball did not alter performance compared to sham cTBS. These findings indicate that SI is critical for extracting somatosensory features (heavy/light) from observed action kinematics and suggest a prominent role of SI in action understanding.
