Abstract/Results: ABSTRACT:
The posterior superior temporal sulcus (pSTS) is a critical node in a network specialized for perceiving emotional facial expressions that is reciprocally connected with early visual cortices (V1/V2). Current models of perceptual decision-making increasingly assign relevance to recursive processing for visual recognition. However, it is unknown whether inducing plasticity into reentrant connections from pSTS to V1/V2 impacts emotion perception. Using a combination of electrophysiological and neurostimulation methods, we demonstrate that strengthening the connectivity from pSTS to V1/V2 selectively increases the ability to perceive facial expressions associated with emotions. This behavior is associated with increased electrophysiological activity in both these brain regions, particularly in V1/V2, and depends on specific temporal parameters of stimulation that follow Hebbian principles. Therefore, we provide evidence that pSTS-to-V1/V2 back-projections are instrumental to perception of emotion from facial stimuli and functionally malleable via manipulation of associative plasticity.

Abstract/Results: ABSTRACT:
Exposure to emotional body postures during perceptual decision-making tasks has been linked to transient suppression of motor reactivity, supporting the monitoring of emotionally relevant information. However, it remains unclear whether this effect occurs implicitly, i.e., when emotional information is irrelevant to the task. To investigate this issue, we used single-pulse transcranial magnetic stimulation (TMS) to assess motor excitability while healthy participants were asked to categorize pictures of body expressions as emotional or neutral (emotion recognition task) or as belonging to a male or a female actor (gender recognition task) while receiving TMS over the motor cortex at 100 and 125 ms after picture onset. Results demonstrated that motor-evoked potentials (MEPs) were reduced for emotional body postures relative to neutral postures during the emotion recognition task. Conversely, MEPs increased for emotional body postures relative to neutral postures during the gender recognition task. These findings indicate that motor inhibition, contingent upon observing emotional body postures, is selectively associated with actively monitoring emotional features. In contrast, observation of emotional body postures prompts motor facilitation when task-relevant features are non-emotional. These findings contribute to embodied cognition models that link emotion perception and action tendencies.

Cortico-cortical paired-associative stimulation (ccPAS) is an advanced dual-site transcranial magnetic stimulation technique that exploits the Hebbian principle to induce plastic changes in functional networks and modulate interactions between cortical brain regions. This review summarizes the growing body of ccPAS research on network dynamics underpinning visual perception. Studies revealed a functional dissociation within cortico-cortical connections in the visual system, where distinct hierarchically organized circuits shape diverse aspects of visual processing, including motion perception, emotion recognition, and metacognitive judgments. Prospective applications integrating ccPAS with neuroimaging techniques such as EEG/MEG hold promise for fine-tuning interventions and gaining deeper insights into visual system network dynamics and functional architecture, with potential clinical applications in neurological and psychiatric conditions.

Efficient inhibitory control in the context of prepotent actions is vital. However, such action inhibition may be profoundly influenced by affective states. Interestingly, research indicates that action control can be either impaired or improved by emotional stimuli. Thus, a great deal of confusion surrounds our knowledge of the complex dynamics subtending emotions and action control. Here, we aimed to investigate whether negative stimuli, even when nonconsciously presented and task-irrelevant, can affect action control relative to neutral stimuli. Additionally, we tested whether individual differences in intracortical excitability may predict action control capabilities. To address these issues, we asked participants to complete a modified version of the Stop Signal Task (SST) in which fearful or neutral stimuli were subliminally presented before the go signals as primes. Moreover, we assessed participants’ resting-state corticospinal excitability, short intracortical inhibition (SICI), and intracortical facilitation (ICF). Results demonstrated better action control capabilities when fearful stimuli were subliminally presented and interindividual SICI predicted stronger action inhibition capabilities. Taken together, these results shed new light on the intricate dynamics between action, consciousness, and motor control, suggesting that intracortical measures can be used as potential biomarkers of reduced motor inhibition in research and clinical settings.

Swiftly halting ongoing motor actions is essential to react to unforeseen and potentially perilous circumstances. However, the neural bases subtending the complex interplay between emotions and motor control have been scarcely investigated. Here, we used an emotional stop signal task (SST) to investigate whether specific neural circuits engaged by action suppression are differently modulated by emotional signals with respect to neutral ones. Participants performed an SST before and after the administration of one session of repetitive transcranial magnetic stimulation (rTMS) over the pre-supplementary motor cortex (pre-SMA), the right inferior frontal gyrus (rIFG), and the left primary motor cortex (lM1). Results show that rTMS over the pre-SMA improved the ability to inhibit prepotent action (i.e., better action control) when emotional stimuli were presented. In contrast, action control in a neutral context was fostered by rTMS over the rIFG. No changes were observed after lM1 stimulation. Intriguingly, individuals with higher impulsivity traits exhibited enhanced motor control when facing neutral stimuli following rIFG stimulation. These results further our understanding of the interplay between emotions and motor functions, shedding light on the selective modulation of neural pathways underpinning these processes.

‘Nip it in the bud’: Low-frequency rTMS of the prefrontal cortex disrupts threat memory consolidation in humans

It is still unclear how the human brain consolidates aversive (e.g., traumatic) memories and whether this process can be disrupted. We hypothesized that the dorsolateral prefrontal cortex (dlPFC) is crucially involved in threat memory consolidation. To test this, we used low-frequency repetitive transcranial magnetic stimulation (LFrTMS) within the memory stabilization time window to disrupt the expression of threat memory. We combined a differential threat-conditioning paradigm with LF-rTMS targeting the dlPFC in the critical condition, and occipital cortex stimulation, delayed dlPFC stimulation, and sham stimulation as control conditions. In the critical condition, defensive reactions to threat were reduced immediately after brain stimulation, and 1 h and 24 h later. In stark contrast, no decrease was observed in the control conditions, thus showing both the anatomical and temporal specificity of our intervention. We provide causal evidence that selectively targeting the dlPFC within
the early consolidation period prevents the persistence and return of conditioned responses. Furthermore, memory disruption lasted longer than the inhibitory window created by our TMS protocol, which suggests that we influenced dlPFC neural activity and hampered the underlying, time-dependent consolidation process. These results provide important insights for future clinical applications aimed at interfering with the consolidation of aversive, threat-related memories.

Decisional confidence refers to the subjective evaluation of the accuracy of a decision based on sensory information. While these judgments are typically grounded in the strength of evidence leading to a decision, they are also subjected to influence from top-down factors such as prior expectations. Previous research has highlighted the impact of prior information on decision parameters such as reaction times and decision criteria placement. However, a comprehensive understanding of how prior information shapes confidence ratings is still lacking. In this study, we manipulate prior knowledge by inducing varying levels of target probability expectation (low: 33%, random: 50%, high: 67%) in a perceptual detection task. In each trial both type-1 (detection) and type-2 (confidence) responses were recorded. First, we replicate previous findings, demonstrating that decisional priors impact decision criteria but not task sensitivity. Secondly, we reveal the strong effect that prior expectations exert on type-2 decisions, with this influence being moderated by a congruency effect between the given prior, the actual stimulus presented, and the provided response. Moreover, we find that confidence is higher in correct compared to incorrect responses, with low-probability trials leading to higher confidence judgments in correct choices compared to random and liberal trials. Finally, we unveil that prior-dependent modulation rates in criterion and confidence were positively associated. These results underscore the intricate interplay between prior expectations, decision-making, and confidence levels, demonstrating that what we perceive is not solely a data-driven phenomenon but can be already shaped by the available information about the state of the world.

Within the realm of schizophrenia, suggested to be a neuropsychiatric disconnection syndrome, discernible aberrations arise in the organization of brain networks. We aim to investigate whether the resting-state functional network is already altered in healthy individuals with high schizotypy traits, highlighting the pivotal influence of brain rhythms in driving brain network alterations.
Study Design
Two-minute resting-state electroencephalography recordings were conducted on healthy participants with low and high schizotypy scores. Subsequently, spectral Granger causality was used to compute functional connectivity in theta, alpha, beta, and gamma frequency bands, and graph theory metrics were employed to assess global and local brain network features.
Study Results
Results highlighted that high-schizotypy individuals exhibit a lower local efficiency in theta and alpha frequencies and a decreased global efficiency across theta, alpha, and beta frequencies. Moreover, high schizotypy is characterized by a lower nodes’ centrality and a frequency-specific decrease of functional connectivity, with a reduced top-down connectivity mostly in slower frequencies and a diminished bottom-up connectivity in faster rhythms.
Conclusions
These results show that healthy individuals with a higher risk of developing psychosis exhibit a less efficient functional brain organization, coupled with a systematic decrease in functional connectivity impacting both bottom-up and top-down processing. These frequency-specific network alterations provide robust support for the dimensional model of schizophrenia, highlighting distinctive neurophysiological signatures in high-schizotypy individuals.

The consolidation process stabilizes a new initially labile memory. This consolidation could operate on a shorter timescale during wakefulness after initial motor learning. Within micro-offline learning states, sequences of simple individual actions learned through interleaved practice are condensed into a unified skill through a time-dependent consolidation process occurring during wakeful periods. While emerging evidence links Glutamate and GABA modulations in the primary motor cortex (M1) to motor learning, its relationship with micro-offline consolidation processes in brief resting states during motor learning is unclear. To investigate this issue, we employed Transcranial magnetic stimulation (TMS) to evaluate whether interindividual variation of different neurotransmitters at rest influences motor learning consolidation in humans. Our results point to the role of GABAB in micro-offline motor consolidation processes during motor learning in M1. This finding could have an important impact on planning neuropharmacology or non-invasive brain stimulation approaches in clinical domains, such as post-stroke rehabilitation.

Objective
The aim of the present study is to examine the relationship between EEG measures and functional recovery in right-hemisphere stroke patients.
Methods
Participants with stroke (PS) and neurologically unimpaired controls (UC) were enrolled. At enrolment, all participants were assessed for motor and cognitive functioning with specific scales (motricity index, trunk control test, Level of Cognitive Functioning, and Functional Independence Measure (FIM). Moreover, EEG data were recorded. At discharge, participants were re-tested with the FIM
Results
Powers in the delta, theta, alpha, and beta bands and connectivity within the fronto-parietal network were compared between groups. Then, the between-group discriminative EEG measures and the motor/cognitive scales were used to feed a machine learning algorithm to predict FIM scores at discharge and the length of hospitalization (LoH). Higher delta, theta, and beta and impaired connectivity were found in PS compared to UC. Moreover, motor/cognitive functioning, beta power, and fronto-parietal connectivity predicted the FIM score at discharge and the LoH (accuracy=73.2 % and 85.2 % respectively).
Conclusions
Results show that the integration of motor/cognitive scales and EEG measures can reveal the rehabilitative potentials of PS predicting their functional outcome and LoH.
Significance
Synergistic clinical and electrophysiological models can support rehabilitative decision-making.

Transcranial magnetic stimulation (TMS) is a widely used tool in the field of clinical and cognitive neuroscience. To exploit its excellent temporal properties, TMS usually relies on triggerbox devices, which temporize the delivery of magnetic pulses according to the paradigm requirements. However, a main limitation of most of the widely used triggerbox devices is that they rely solely on the experimental computer processor, which might add temporal uncertainty in delivering the TMS pulse when the computer’s resources are drained by other experimental devices or by task execution itself, especially during repetitive TMS or dual-coil protocols. We aimed at developing a low-cost and easily reproducible triggerbox device which could overcome these limitations by relying on an external processor to handle the timing precision. We used an Arduino Uno R4 Minima to build Silicon Spike, a low-cost ($60) triggerbox device. We tested the device’s precision in delivering the TMS pulses under different working load conditions, and the impact over time. All of the tests were ecological, delivering real TMS pulses during dual-coil, repetitive, and patterned TMS protocols. We obtained extremely high precision (< 0.022 ms) in all of the tests. This means that, for smaller or longer latencies, the error remains negligible for TMS studies. Thus, the Silicon Spike device demonstrated microsecond precision in handling the TMS pulse delivery, establishing itself as a simple and yet precise device. We freely provide the source code and the hardware schematics, allowing anyone to reproduce our work.

ABSTRACT:
Background
Effective inhibitory control over prepotent actions is crucial. Understanding the brain mechanisms that enable humans to suppress unwanted behaviors remains a major scientific challenge that has been relatively underexplored. Notably, inhibitory control appears to be affected by the presence of emotional stimuli, although findings in this area have been mixed.
Aims
We aim to disclose the complex interplay between action control and emotions, investigating how emotional stimuli are able to behaviorally influence action control as well as the neural dynamics subtending such complex executive function.
Method
In a series of behavioral and Transcranial Magnetic Stimulation (TMS) studies, we asked healthy young participants to complete action control tasks. We investigated how action control capabilities can be ameliorated or suppressed due to different TMS protocol applications and whether individual neurophysiological indices may predict action suppression abilities.
Results
We demonstrated that negative emotions are able to influence action control and that the neural dynamics subtending action suppression are differently influenced in a neutral vs an emotional context. Interestingly, individual differences in intracortical mechanisms within the motor system may be considered as a biomarker for action control capabilities.
Conclusions
This project significantly advances the mechanistic understanding of the complex interplay between action and emotion, demonstrating that negative emotions can ameliorate action suppression. Taken together, our results pave the way for future TMS-based treatments in several psychiatric and clinical disorders with serious deficits in action control, especially in emotion-related context.

Abstract/Results: ABSTRACT:
The spectrum of schizophrenia is characterised by an altered sense of self with known impairments in tactile sensitivity, proprioception, body-self boundaries, and self-recognition. These are thought to be produced by failures in multisensory integration mechanisms, commonly observed as enlarged temporal binding windows during audiovisual illusion tasks. To our knowledge, there is an absence of computational explanations for multisensory integration deficits in patients with schizophrenia and individuals with high schizotypy, particularly at the neurobiological level. We implemented a multisensory causal inference network to reproduce the responses of individuals who scored low in schizotypy in a simulated double flash illusion task. Next, we explored the effects of recurrent excitation, cross-modal and feedback weights, and synaptic density on the visual illusory responses of the network. Using quantitative fitting to empirical data, we found that an increase in the weights of the recurrent excitatory connectivity in the network enlarges the temporal binding window and increases the overall proneness to experience the illusion, matching the responses of individuals scoring high in schizotypy. Moreover, we found that an increase in excitation increases the probability of inferring a common cause from the stimuli. We propose an E/I imbalance account of reduced temporal discrimination in the SCZ spectrum and discuss possible links with Bayesian theories of schizophrenia. We highlight the importance of adopting a multisensory causal inference perspective to address body-related symptomatology of schizophrenia.

Abstract/Results: ABSTRACT:
Fearful body expressions convey critical information that is rapidly and preferentially processed, facilitating swift motor reactions to potential dangers. Consistent evidence has shown that even the subliminal presentation of fear-related expressions can impact visual processing and autonomic responses, increasing sensory vigilance for monitoring potential threats. However, it remains unclear whether the presentation of non-visible emotional bodies modulates corticospinal excitability (CSE) in the observer. To investigate this, we asked 22 healthy participants to perform a sex discrimination task involving neutral target body postures, preceded by the brief subliminal presentation of fearful, happy, or neutral body postures. CSE was tested using Transcranial Magnetic Stimulation (TMS) at early time points (70, 90, and 110 ms) after target stimulus onset. Results showed a significant CSE reduction in the dominant hemisphere for subliminal fearful primes compared to happy and neutral primes. This CSE suppression was independent of the time of stimulation, participants’ subjective or objective awareness, metacognitive sensitivity, or personality traits. Our findings highlight an early automatic activation of the motor system in response to subliminal fearful stimuli, supporting the view that fearful expressions, even when not consciously perceived, activate basic survival mechanisms for monitoring and preparing fast motor responses to potential threats.

Perception relies on hierarchical processes integrating sensory data into higher-order models about the world. In the sensory domain, this hierarchy also involves horizontal pathways aiding interhemispheric interactions. For example, recent focus on the V5-V5 network revealed its role in motion processing. However, despite converging evidence demonstrating the crucial role of oscillatory activity in inter-areas communication, the specific rhythmic code governing V5-V5 interaction is unclear. Through traveling wave analysis, this gap is filled by identifying the alpha/low-beta frequency (9–16 Hz) as crucial for long-range, interhemispheric communication between V5 regions. Notably, a pronounced imbalance is observed: stronger waves travelled in the left-to-right-V5 direction. Crucially, this observation extends to the behavioural level, as Signal Detection Theory analysis uncovered a correlation between the highlighted imbalance and a perceptual bias favouring leftward motion reporting. Our findings underscore asymmetrical projections between V5 regions, highlighting the interplay between inter-hemispheric traveling wave patterns in dictating motion perception.

Abstract/Results: ABSTRACT:
Perceiving the duration of events is a fundamental ability for everyday life. Traditional research has focused on the role of alpha oscillations as an endogenous pacemaker for the human internal clock, yet there is limited evidence supporting this idea. An alternative hypothesis proposes that alpha oscillations may underlie a sampling mechanism, where higher alpha frequencies correspond to increased information sampling, resulting in more accurate temporal judgments. In this study, we tested the internal clock versus sampling rate hypothesis by examining the relationship between Individual Alpha Frequency (IAF) and fine-grained time perception. Using resting Electroencephalography (EEG) and Signal Detection Theory (SDT), fifty healthy volunteers performed a time-discrimination task with 100 and 500 ms standard durations. Our results demonstrate that temporal sensitivity (d’) but not temporal bias (c) is influenced by IAF, with higher IAF leading to more accurate time estimates (higher d’). The correlations were observed over frontocentral topographies consistent with previous reports of neural networks involved in time processing and were most pronounced at 100 ms relative to 500 ms, likely due to fluctuations in IAF across multiple cycles. In conclusion, our findings support the relationship between IAF and temporal sensitivity. These results challenge the pacemaker hypothesis and instead suggest a distributed mechanism where alpha oscillations enhance the precision of temporal sampling. Our study adds to the growing body of evidence highlighting the role of IAF in sensory sampling as a generative mechanism for temporal sensitivity as opposed to subjective time perception.

The BIAL Foundation was created in 1994 by Laboratórios BIAL in conjunction with the Council of Rectors of Portuguese Universities. BIAL’s Foundation mission is to foster the scientific study of Man from both the physical and spiritual perspectives.

Reactive inhibition is crucial for preventing inappropriate actions, and impairments in this ability are common in various disorders, with the underlying neural mechanisms poorly understood. To explore the neural dynamics of such an ability, we used a novel transcranial magnetic stimulation (TMS) protocol, cortico-cortical paired associative stimulation (ccPAS), to induce Hebbian spike-timing-dependent plasticity (STDP). Our goal was to investigate the functional relevance of key brain regions in the action inhibition network (AIN). Healthy participants underwent ccPAS targeting functional connectivity between AIN regions: pre/supplementary motor area (preSMA/SMA) to the left motor cortex (lM1), right inferior frontal gyrus (rIFG) to lM1, or right M1 (rM1) to lM1. Participants completed a stop signal task (SST) before and after ccPAS stimulation. Motor evoked potentials (MEP) were recorded during ccPAS to assess network plasticity, and resting motor threshold (rMT) was measured for global motor excitability. Reactive inhibition improved selectively in the preSMA/SMA-lM1 group, with MEPs increasing after preSMA/SMA-lM1 and rM1-lM1 stimulation, suggesting facilitatory modulations. rMT correlated with behavioral improvement in the preSMA/SMA group. These findings demonstrate that ccPAS improved reactive inhibition, enhancing plasticity between preSMA/SMA and lM1, providing insights into the AIN's functional mechanism.
